摘  要:该文开展了青钱柳降糖降尿酸活性部位的含量测定和指纹图谱研究,评价了不同产地青钱柳药材的质量。在活性筛选的指导下优化提取条件,以30%乙醇水溶液为提取溶剂,料液比1∶10,超声提取1 h可有效提取活性部位。采用高效液相色谱法测定绿原酸、槲皮素-3-O-β-D-葡萄糖醛酸苷、槲皮苷、山奈酚-3-O-β-D-葡萄糖醛酸苷、异绿原酸C和阿福豆苷的含量,从各成分含量随着产地的波动情况比较青钱柳药材的质量,其中江西药材S1的成分含量较低,与其它产地药材差异较大。同时建立了青钱柳活性部位的指纹图谱,10批药材的相似度均大于0.9,主成分分析(PCA)表明,江西药材S1、湖南药材S6和S8质量差异较大。10个产地药材的α-糖苷酶和黄嘌呤氧化酶活性抑制实验表明,湖南药材S5的降糖和降尿酸活性最佳,而江西药材S1表现较差,与质量评价结果基本一致。所建方法可为青钱柳的质量控制和质量标准建立提供科学依据。This paper carried out the content determination and fingerprinting study of the hypoglyce⁃mic and uric acid-lowering active parts of Cyclocarya paliurus.(C.paliurus.),and evaluated the quality of C.paliurus.herbs from different origins.Under the guidance of activity screening,the ex⁃traction conditions were optimized.Using 30%ethanol aqueous solution as extraction solvent,a sol⁃id-liquid ratio of 1∶10,and ultrasonic extraction for 1 hour can extract the active part effectively.A high performance liquid chromatography(HPLC)method was used for the determination of the con⁃tents of chlorogenic acid,quercetin-3-O-β-D-glucuronide,quercitrin,kaempferol-3-O-β-D-glucuro⁃nide,isochlorogenic acid C and afzelin in the active part of C.paliurus..Quality evaluation was based on the content variation of each ingredients with the origin.Among them,the Jiangxi herb S1 had the low content,which differed from the herbs of other origins.At the same time,a fingerprint of the active part of C.paliurus.was established.The similarity of the 10 batches of herbs was all greater than 0.9.PCA analysis showed that the quality differences of Jiangxi herb S1,Hunan herbs S6 and S8 were large.The inhibitory activities ofα-glucosidase and xanthine oxidase showed that Hu⁃nan herb S5 had the best hypoglycemic and uric acid-lowering activities,while Jiangxi herb S1 per⁃formed poorly,providing consistent conclusions with quality evaluation.The established method pro⁃vide a scientific basis for the quality control and the quality standard of C.paliurus.herb.
