摘 要:以白桦叶片cDNA为模板使用RT-PCR技术克隆BpEIN3基因,并通过双酶切成功构建pBI121-BpEIN3-EGFP表达质粒,使用农杆菌转化法获得BpEIN3白桦过表达株系,并研究过表达BpEIN3基因对白桦形态学的影响及盐胁迫后对生理生化的影响。结果表明:与野生型白桦相比,同环境下生长的4月生的转BpEIN3基因幼苗株高有所降低。盐胁迫下,BpEIN3过表达植株叶片叶绿素质量分数和超氧化物歧化酶(SOD)活性显著高于野生型,丙二醛(MDA)质量摩尔浓度和相对电导率则显著低于野生型。说明白桦BpEIN3基因在耐盐方面发挥着重要作用。The BpEIN3 gene was cloned using RT-PCR with Betula platyphylla leaf cDNA as a template,and the pBI121-BpEIN3-EGFP expression plasmid was successfully constructed through double enzyme digestion.BpEIN3 B.platyphylla overexpression lines were obtained using Agrobacterium⁃mediated transformation,and the effects of overexpressed BpEIN3 gene on the morphology of B.platyphylla and its physiological and biochemical changes under salt stress were studied.The results showed that compared to the wild⁃type B.platyphylla,the height of the 4⁃month⁃old transgenic seedlings overex⁃pressing BpEIN3 gene was significantly decreased under the same environmental conditions.Under salt stress,the trans⁃genic plants overexpressing BpEIN3 gene showed significantly higher chlorophyll content and superoxide dismutase(SOD)activity compared to the wild⁃type,while the malondialdehyde(MDA)quality molar concentration and relative conductivi⁃ty were significantly lower than the wild⁃type.These results showed that the BpEIN3 gene plays an important role in salt tol⁃erance in B.platyphylla.
