摘  要:【目的】为了预测甜菜锌指蛋白BvZAT10(LOC 104901462)的结构特点和功能。【方法】以甜菜‘780016B/12优’为试验材料,对BvZAT10基因进行了克隆及生物信息学分析,并利用甜菜响应镉胁迫的转录表达谱数据探究了该基因在镉胁迫下的应答特性。【结果】BvZAT10编码区全长729 bp,编码242个氨基酸,是一种碱性不稳定亲水性蛋白,具有2个C2H2型锌指结构域。该基因编码蛋白共有36处磷酸化修饰位点,蛋白结构以无规则卷曲为主,系统发育分析发现BvZAT10与菠菜和藜麦的ZAT10亲缘关系最近。BvZAT10启动子序列包含植物激素响应、光响应及抗逆相关的多种作用元件。在1、3、5 mmol/L CdCl_(2)处理下,BvZAT10及其预测调控的大部分靶基因,如金属耐受蛋白11(BVRB_2g037080)和ABCG家族成员22(BVRB_5g109050)等,均受到了镉胁迫的正向调控。【结论】推测BvZAT10可能直接或间接地在甜菜应答镉胁迫中发挥重要作用,可将其作为潜在优势抗逆基因深入开展后续研究。【Objective】To predict the structural characteristics and functions of sugar beet zinc finger protein BvZAT10(LOC 104901462),【Methods】using sugar beet‘780016B/12 superior’as experimental material,BvZAT10 gene was cloned and subjected to bioinformatics analysis,and the transcriptional expression profile data of sugar beet in response to cadmium stress were used to explore the gene response characteristics under cadmium stress.【Results】The coding region of BvZAT10 is 729 bp in length,encoding 242 amino acids.It is an alkaline,unstable hydrophilic protein with two C2H2-type zinc finger domains.The protein encoded by this gene has a total of 36 phosphorylation modification sites,and the protein structure is mainly characterized by irregular coiling.Phylogenetic analysis revealed that BvZAT10 is most closely related to ZAT10 in spinach and quinoa.The BvZAT10 promoter sequence contains multiple functional elements related to plant hormone response,light response,and stress resistance.Under 1,3 and 5 mmol/L CdCl_(2) treatments,BvZAT10 and most of its predicted regulatory target genes,such as metal tolerance protein 11(BVRB_2g037080)and ABCG family member 22(BVRB_5g109050),were positively regulated by cadmium stress.【Conclusion】It is speculated that BvZAT10 may play an important role directly or indirectly in sugar beets response to cadmium stress and could be further studied as a potential dominant stress-tolerant gene.
