王族海棠Alfin-like家族基因的鉴定及盐胁迫下的表达分析

摘  要:Alfin-like(AL)转录因子家族对非生物胁迫反应具有重要的调控作用。本研究采用同源比对的方法检索鉴定王族海棠AL转录因子家族基因,系统分析其生物信息学特性,并采用qRT-PCR方法分析其在盐胁迫下的表达模式,以期为揭示AL家族在王族海棠响应盐胁迫中的生理功能提供理论依据。结果表明,从王族海棠基因组中共鉴定出11个MdALs基因,分布在6条染色体上,分别命名为MdAL1—MdAL11。MdALs转录因子具有高度保守的Alfin结构域和PHD锌指结构域,含4~10个内含子;上游启动子区域有大量与植物激素和非生物胁迫响应相关的顺式作用元件,且基因片段复制事件在MdAL基因家族的扩展中起着至关重要的作用。转录组分析显示MdALs基因主要在王族海棠生育后期高表达。qRT-PCR分析进一步表明,MdALs基因在王族海棠遭受盐胁迫下的表达模式不同,盐胁迫下,MdAL3、MdAL4、MdAL6基因的表达量整体上调,尤其MdAL4基因,其表达水平随着盐胁迫处理时间的延长呈显著增加趋势。综上,王族海棠AL转录因子家族与植物响应激素变化和非生物胁迫密切相关,尤其是MdAL4基因,强烈响应盐胁迫,这可为利用基因工程技术改良王族海棠种质奠定基础。The Alfin⁃like(AL)transcription factor family play important regulatory roles in response to abiotic stresses.In this study,the homologous alignment method was used to search and identify the AL tran⁃scription factor family genes of Malus doumeri‘Royalty’,the bioinformatics characteristics were systematically analyzed,and their expression patterns under salt stress were analyzed by qRT⁃PCR method in order to provide a theoretical basis for revealing the physiological function of AL family in response to salt stress.The results showed that 11 MdAL genes were identified from the genome of M.doumeri‘Royalty’,which were distributed on 6 chromosomes and named from MdAL1 to MdAL11,respectively.The MdAL proteins had highly conserved Alfin domain and PHD zinc finger domain,containing 4~10 introns.The upstream promoter region contained a large number of cis⁃acting elements relating to plant hormone and abiotic stress responses,and gene fragment replication events played a crucial role in the expansion of the MdAL gene family.Transcriptome analysis showed that MdALs were mainly expressed highly in late growth period of M.doumeri‘Royalty’.Furthermore,the expression patterns of the MdAL genes were different under salt stress.The expression level of MdAL3,MdAL4 and MdAL6 genes were up⁃regulated under salt stress,especially MdAL4,whose expression level in⁃creased significantly with the prolongation of salt stress time.In summary,the AL transcription factor family of M.doumeri‘Royalty’was closely related to plant hormone and abiotic stresses,especially the MdAL4 gene,which strongly responded to salt stress.All these results could lay the foundation for improving M.doumeri‘Royalty’germplasms using genetic engineering technologies.
