摘 要:研究旨在建立不同冷凝回流提取条件下黑沙蒿醇提物甲醇溶解的高效液相色谱法(HPLC)指纹图谱,对其药效指标和指纹图谱的相关性进行分析,以期进一步揭示黑沙蒿醇提物起抑菌效果的药效物质。通过冷凝回流提取获得黑沙蒿醇提物,使用HPLC建立黑沙蒿醇提物醇溶HPLC指纹图谱,共标记10个共有峰,指纹图谱相似度均在0.979~1.000范围内,拥有良好的稳定性,并对得到的黑沙蒿醇提物进行体外抑菌试验,体外抑菌试验表现出对金黄色葡萄球菌的抑菌效果,结合指纹图谱及抑菌试验结果进行谱效关系分析,通过灰色关联度分析起作用的共有峰。结果表明,抑菌效果最强的提取条件为乙醇浓度65%,水浴温度95℃,料液比1∶9,水浴时间2 h,且各提取条件下提取的成分基本相似。10个共有峰全部与抑菌效果关联度大于0.6,且其中有4个峰与抑菌效果关联度大于0.8。黑沙蒿醇提物的抑菌效果是多成分协同作用的结果。通过指纹图谱与抑菌效果的相关性初步明确了黑沙蒿醇提物抑菌活性成分的有效峰,筛选出了黑沙蒿醇提物中发挥抑菌作用的有效峰群,为黑沙蒿醇提物的抑菌物质基础研究及应用提供参考。This study aimed to establish the HPLC fingerprints of Artemisia Ordosica Krasch.annua alcohol extract under different condensation reflux extraction conditions in water and methanol solubilization conditions,and to analyze the correlation between the pharmacodynamic indexes and the fingerprints to further reveal the pharmacodynamic substances of Artemisia Ordosica Krasch.alcoholic extracts.Artemisia Ordosica Krasch.extracts were obtained by condensation reflux extraction,and the HPLC fingerprint of Artemisia Ordosica Krasch.extracts in alcohol solution was established by HPLC.A total of 10 common peaks were marked,and the similarity of the fingerprint was between 0.979and 1.000,with good stability.In vitro antibacterial test was carried out on the obtained Artemisia Ordosica Krasch.extracts,and the in vitro antibacterial test showed the antibacterial effect on Staphylococcus aureus.The spectral-effective relationship was analyzed by combining the fingerprint and the results of the antibacterial test,and the common peaks playing a role were analyzed by gray correlation.The results showed that the optimal extraction conditions were as follows:ethanol concentration of 65%,water bath temperature of 95 ℃,solid-liquid ratio of 1:9,water bath time of 2 hours.The extracted components exhibited similar profiles under each extraction condition.Moreover,a strong correlation(greater than 0.6) was observed between the bacteriostatic effect and ten common peaks,while four peaks showed an even higher correlation(greater than 0.8).Therefore,the antibacterial effect of Artemisia Ordosica Krasch.extracts was the result of the synergistic effect of multiple components.The effective peaks of the antibacterial active components of Artemisia Ordosica Krasch.extracts were preliminarily determined by the correlation between the fingerprint and the antibacterial effect,and the effective peaks of Artemisia Ordosica Krasch.extracts with antibacterial effect were screened out,which provided a reference for the basic research and applicat
