摘　　要：目的 探讨外泌体miR-146a-5p、miR-155-5p在寻常型银屑病患者血浆、唾液、尿液中的表达及意义。方法 采用超速离心法提取40例寻常型银屑病患者血浆、唾液和尿液中外泌体,利用投射电镜观察外泌体形态,并提取RNA,利用核酸测定仪检测RNA浓度,RT-qPCR检测miR-146a-5p、miR-155-5p的表达水平。结果 外泌体外形呈现圆形或椭圆形,直径均在60~200 nm;外泌体RNA在银屑病患者血浆、唾液、尿液中的浓度分别为3.9 ng/μl、61.6 ng/μl、0.57 ng/μl;外泌体miR-146a-5p、miR-155-5p在寻常型银屑病患者血浆、唾液、尿液中的表达分别为血浆(0.01±0.002)、(0.06±0.002),尿液(1.03±0.02)、(0.95±0.08),唾液(0.02±0.00)、(0.18±0.01)。结论 外泌体miR-146a-5p、miR-155-5p在寻常型银屑病患者不同体液中存在表达性差异,从微观角度研究银屑病,有利于为今后的诊断、治疗及发病机制的研究提供科研依据。Objective To investigate the expression and significance of exosome miR-146a-5p and miR-155-5p in plasma,saliva and urine of psoriasis vulgaris.Methods Exosomes in plasma,saliva and urine of 40 patients with psoriasis vulgaris were extracted by ultracentrifugation,exosome morphology was observed by projection electron microscopy,RNA was extracted,RNA concentrationwas detected by nucleic acid analyzer,and the expression levels of miR-146a-5p and miR-155-5p were detected by RT-qPCR.Results The shape of exosomes was round or oval,and the diameter was 60~200 nm.The concentrations of exosomal RNA in plasma,saliva and urine of psoriasis patients were 3.9 ng/μl,61.6 ng/μl and 0.57 ng/μl,respectively.The expressions of exosome miR-146a-5p and miR-155-5p in plasma,saliva and urine of patients with psoriasis vulgaris were plasma(0.01±0.002),(0.06±0.002),urine(1.03±0.02),(0.95±0.08),saliva(0.02±0.00),(0.18±0.01),respectively.Conclusion There are expressive differences between exosomes miR-146a-5p and miR-155-5p in different body fluids of patients with psoriasis vulgaris,and the study of psoriasis from a microscopic perspective is conducive to providing scientific research basis for future diagnosis,treatment and pathogenesis.
