摘 要:污蝇杆菌(Wohlfahrtiimonas chitiniclastica)是一类新出现的罕见人畜共患致病菌,可侵袭动物机体并引起化脓性病症。2018年贵州某猪场送检了2只患病仔猪,呈典型的副猪嗜血杆菌(H.parasuis)混合感染病症。无菌采集病猪肺脏组织,进行细菌培养,通过16S rRNA测序检测到污蝇杆菌。为了解该病原特性,对该菌进行分离纯化培养。从培养特性、革兰染色、生化试验、16S rRNA序列分析、电镜观察等方面对该菌进行系统鉴定;采用药敏试验测定其药物敏感性;通过人工感染小鼠确定其致病性。结果显示,该分离菌株的16S rRNA基因与污蝇杆菌参考株核苷酸相似性介于95.9%~97.6%,与参考株KBL006(NR_133893.1)遗传距离最近;分离株在血平板、LB固体培养基等的生长良好,在高盐培养基上不生长;生化鉴定结果显示分离菌株的生化特性与污蝇杆菌符合;扫描电镜观察显示该菌菌体长宽为1.30μm×0.53μm;药敏试验结果显示分离株对头孢菌素类、多黏菌素B及部分青霉素、氨基糖苷类药物较为敏感,对四环素类、大环内酯类及喹诺酮类等药物耐药;小鼠致病性试验结果显示分离菌株对小鼠具有致死能力。本试验成功分离到1株猪源污蝇杆菌并对其病原生物学特性进行了初步研究,结果为污蝇杆菌病的病原检测和防控治疗提供了重要参考依据。Wohlfahrtiimonas chitinicolatica(W.chitinicolatica)is a rare newly emerged zoonotic pathogen that can invade animal bodies and cause purulent diseases.In 2018,a pig farm in Guizhou sent two sick piglets for testing,which showed a typical H.parasuis mixed infection.Sterile collection of diseased pig lung tissue was cultured,W.chitinicolatica was detected by 16S rRNA sequencing.To understand the pathogenic characteristics,the pathogen was isolated,purified and cultured.The pathogen was systematically identified according to culture characteristics of Gram stain,biochemical test,16S rRNA sequence analysis,and electron microscopy observation.Its drug susceptibility was determined through susceptibility testing;its pathogenicity was determined by the mouse pathogenicity test.The results showed that the nucleotide similarity between the isolate and W.chitinicolatica was 95.9%-97.6%,which is the closest genetic distance from the reference strain KBL006(NR_133893.1).It grows well on blood agar plates,LB agar medium,etc.,but does not grow on high-salt medium;biochemical identification results showed that the biochemical characteristics of the isolates were consistent with W.chitinicolatica.Electron microscopy showed that the length and width of the bacteria were 1.30μm×0.53μm;the results of drug susceptibility test showed that the isolates were sensitive to cephalosporins,polymyxin B,certain penicillins and aminoglycosides,but resistant to tetracyclines,macrolides,and quinolones.The results of the mouse pathogenicity test showed that the isolated strain was lethal to mice.In this study,a strain of W.chitinicolatica sucis was successfully isolated,and its pathogenic biological characteristics were preliminarily studied,and the results provide an important reference for the pathogen detection,prevention and control of W.chitinicolatica.
