摘 要:产气荚膜梭菌(Clostridium perfringens)可引起人类食物中毒和许多非食源性人类胃肠道(GI)疾病。编码其肠毒素(Clostridium perfringens enterotoxin, CPE)的基因cpe若位于染色体的菌易引起食物中毒,cpe位于质粒的菌与非食源性GI疾病相关,因此本研究应用PCR检测、单位点序列分型分析(SLST)、耐热性测定,综合分析cpe+菌的cpe定位。结果显示,双重PCR检测29株cpe+菌仅扩增出600 bp的cpe目的条带,未检测出IS1470、IS1470-like、IS1151序列;通过sod、sodFPF PCR检测及SLST分析推测所有检测菌的cpe可能均位于染色体上;7-1和21-3的活菌及芽胞耐热性均高于42-2,推测两者cpe可能位于染色体,而42-2 cpe可能位于质粒上。结果为产气荚膜梭菌引起食物中毒和非食源性疾病的诊断和防控以及cpe的位置与所致疾病相关性的研究提供材料和科学依据。Clostridium perfringens(C.perfringens)can cause food poisoning in humans and many non-foodborne gastrointestinal(GI)diseases.C.perfringens enterotoxin(CPE),whose encoding gene cpe locates on the chromosome is prone to cause human food poisoning,while C.perfringens with cpe locates on the plasmid is associated with non-foodborne human GI diseases.In this study,the localization of cpe of cpe~(+)isolates were analyzed by PCR amplification,single-locus sequence typing(SLST)as well as the heat resistance of vegetative cells and spores.The duplex PCR results showed that only 600 bp bands were amplified from 29 cpe~+isolates,IS1470,IS1470-like and IS1151 target bands were not detected.The conventional sod and sodFPF PCR detection as well as SLST analysis showed that cpe of the detected isolates may locate on chromosome.The heat resistance of vegetative cells and spores of strain 7-1 and 21-3 were higher than that of 42-2,it was speculated that cpe of 7-1 and 21-3 locate on chromosomes while cpe of 42-2 locates on plasmid.In conclusion,it provides materials and scientific basis for the diagnosis,prevention and control of food poisoning and non-foodborne diseases,as well as the correlation between cpe location and the disease caused by C.perfringens.
