摘 要:在室温条件下制备具有过氧化物酶样活性的WO_(3)纳米片(WO_(3)NSs),基于WO_(3)NSs催化H_(2)O_(2)氧化3,3',5,5'-四甲基联苯胺(TMB)生成蓝色氧化产物(ox-TMB)及在尿酸氧化酶催化下,尿酸被氧化并定量释放H_(2)O_(2)的原理,建立一种以WO_(3)NSs为过氧化物模拟酶的尿酸测定方法。考察了多种因素对吸光度响应的影响。在优化的实验条件下,当尿酸浓度在1.0~100μmol/L范围内时,吸光度与浓度呈良好的线性关系,方法的检出限为0.61μmol/L,应用于血清样品中尿酸的测定,回收率在97.1%~101.4%范围内。WO_(3) nanosheets(WO_(3)NSs)with peroxidase-like activity were prepared at room temperature,which could catalyze the oxidation of 3,3',5,5'-tetramethylbenzidine(TMB)by H_(2)O_(2) to produce a blue oxidation product(ox-TMB).Under the catalysis of uric acid oxidase,uric acid is oxidized and H_(2)O_(2) was quantitatively released.Based on of these,a method for the determination of uric acid using WO_(3)NSs as a peroxidase mimic enzyme was established.The effects of various factors on absorbance response were investigated.Under optimized experimental conditions,there was a good linear relationship between absorbance and uric acid concentration in the range of 1.0 to 100μmol/L with a detection limit of 0.61μmol/L.The method exhibits its potential application in the detection of uric acid in serum samples with the recovery of 97.1%to 101.4%.
