摘 要:目的在细胞水平观察桑藿排毒茶对甲型H1N1流感病毒的抑制作用。方法利用CCK-8试剂检测桑藿排毒茶的细胞毒性,通过分析不同处理组中甲型流感病毒RNA与蛋白表达水平的变化,评价该组方抗甲型流感病毒活性的高低。结果桑藿排毒茶与磷酸奥司他韦对犬肾细胞(MDCK)的最大无毒浓度(TC 0)分别为4.32、1.02 mg·mL^(-1),半数有效浓度(EC 50)分别为0.38 mg·mL^(-1)、3.5μg·mL^(-1)。在Q-PCR与Western blotting试验中,相较于病毒组,经药物处理后的感染细胞中流感病毒RNA与蛋白的表达量均显著降低,说明其对病毒的抑制作用较强。结论桑藿排毒茶在细胞水平可以抵抗甲型流感病毒感染,具有进一步开发意义。Objective To observe the inhibitory effect of Sanghuo Paidu Tea on influenza A(H1N1)virus at the cellular level.Methods CCK-8 reagent was used to detect the cytotoxicity of Sanghuo Paidu Tea,and the anti-influenza A virus activity of Sanghuo Paidu Tea was evaluated by analyzing the changes in the expression levels of influenza A virus RNA and protein in different treatment groups.Results The maximal atoxic concentration(TC 0)of Sanghuo Paidu Tea and Oseltamivir phosphate for MDCK was 4.32 mg·mL^(-1) and 1.02 mg·mL^(-1),respectively,and the 50%effective concentration(EC 50)was 0.38 mg·mL^(-1) and 3.5μg·mL^(-1).In Q-PCR and Western blotting experiments,compared to the viral group,the expression levels of influenza virus RNA and protein in infected cells treated with drugs were significantly reduced,indicating a strong inhibitory effect on the virus.Conclusion Sanghuo Paidu Tea can resist the infection of influenza A virus at the cellular level,and has further development significance.
