摘 要:目的建立白头翁药材的超高效液相色谱(UPLC)特征图谱方法,为白头翁药材的产地区分及质量控制提供参考。方法采用UPLC法,色谱柱为Agilent SB C18(2.1 mm×100 mm,1.8μm);以乙腈-甲醇(2∶1)-0.1%磷酸溶液为流动相,梯度洗脱;流速0.30 ml/min;柱温30℃;检测波长215 nm;进样量2μl。通过特征图谱对比及主成分分析、正交偏最小二乘判别分析(OPLS-DA)对常见的白头翁伪品及不同产地的白头翁药材进行评价。结果白头翁药材特征图谱呈现9个特征峰,通过高分辨质谱及对照品对比,指认了其中8个共有峰。通过主成分分析,可明显区分不同产地的白头翁药材,并结合OPLS-DA分析发现,峰2、峰3、峰6是不同产地白头翁差异的主要标志物。结论所建立的方法专属性、重复性、耐用性良好,可有效区分白头翁的常见伪品,并为白头翁药材的质量控制及产地选择提供依据。Objective To establish ultra performance liquid chromatography(UPLC)characteristic chromatogram of Pulsatilla chinensis;To provide reference for the origin identification and quality control of Pulsatilla chinensis.Methods UPLC Method was adopted.The determination was performed on a column of Agilent SB C18(2.1 mm×100 mm,1.8μm).The mobile phase was acetonitrile-methanol(2:1)-0.1%phosphoric acid solution by fradient elution at a flow rate of 0.30ml/min.The column temperature was 30℃.The detection wavelength was 215 nm.The injection volume was 2μl.The common counterfeit products and medicinal herbs of Pulsatilla chinensis from different areas were evaluated by comparison of characteristic chromatogram,principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA).Results There were 9 characteristic peaks in the characteristic chromatogram of Pulsatilla chinensis,and 8 common peaks were identified by high resolution mass spectrometry and comparison of reference materials.Through PCA analysis,it was possible to clearly distinguish the medicinal herbs of Pulsatilla chinensis from different areas.Combined with OPLS-DA analysis,it was found that peak 2,peak 3,peak 6 were the main markers of Pulsatilla chinensis from different producing areas.Conclusion The established method has good specificity,repeatability and durability,and it can effectively distinguish the common counterfeits of Pulsatilla chinensis,and provide the basis of quality control and selection of origin for Pulsatilla chinensis.
