摘  要:以黑木耳“吉蕈-A8”为试材,对其进行继代培养,研究继代次数对菌丝生长特性、显微特征、胞外酶活性、遗传变异及农艺性状等菌种活力指标的影响,以期为菌种退化与继代次数相关性研究提供参考依据。结果表明:菌种在继代15次内能保持遗传稳定性;但继代培养会使菌丝变得更加细长,分支减少;F_(1)~F_(7)区间整体活力高,在F7代左右生长速率更快,漆酶、淀粉酶和蛋白酶活性能维持较高水平,菌丝尖端粗壮,分支多且密,与最终农艺性状呈正相关。在生产中要确保黑木耳菌种高活力,继代次数应控制在3~6次。Taking Auricularia heimuer‘Jixun-A8’as the test material,it was subcultured to study the effect of subculture times on the mycelial growth characteristics,microscopic characteristics,extracellular enzyme activity,genetic variation and agronomic traits of the strain,in order to provide reference for the study of the correlation between strain degradation and subculture times.The results showed that the strain could maintain genetic stability within 15 generations;however,subculture would make the mycelia more slender and the branches fewer.The overall vitality was high in the F_(1)-F_(7)interval,with a faster growth rate around the F_(7)generation,higher laccase,amylase and protease activities,thicker and denser hyphal tips,and more branches,which were positively correlated with the final agronomic traits.In production,to ensure the high vitality of Auricularia heimuer strains,the number of subcultures should be controlled within 3-6 times.
