摘  要:近年来体外替代实验在胃肠道毒性研究中愈发重要,传统的新药非临床研究主要以动物实验为主,但随着生物技术的发展和动物福利的要求,胃肠道模型已从体内实验逐渐转向体外,从单一的细胞类型过渡到更加接近体内环境的类器官共培养模型,甚至可以进行多器官融合培养和微流控培养等。这些培养体系通过模拟与体内一致的生理和病理环境,既可以用于胃肠道疾病建模,也可作为药物毒性筛选平台促进体外毒性评价研究进展。本文主要综述了常规体外胃肠道模型、胃肠道类器官模型和微流控芯片的研究进展,以期促进体外研究发展。In recent years,in vitro substitution tests have become increasingly important in gastrointestinal toxicity studies.Traditional non-clinical studies of new drugs are mainly based on animal experiments.However,with the development of biotechnology and the requirements of animal welfare,in vitro gastrointestinal model has gradually shifted from in vivo tests to in vitro,from a single cell type to an organoid co-culture model that is closer to the in vivo environment.It can even be used for multi-organ fusion culture and microfluidic culture.By simulating the physiological and pathological environment consistent with in vivo,these culture systems can be used not only for the modeling of gastrointestinal diseases,but also as a platform for drug toxicity screening,which promotes the progress of in vitro toxicity evaluation.In this paper,the research progress of conventional in vitro gastrointestinal model,gastrointestinal organoid model and microfluidic chip are reviewed.
