摘 要:为评价菊苣酸改善猪德尔塔冠状病毒(PDCoV)感染仔猪后引起其的肠道菌群失衡,本研究设置菊苣酸组、菊苣酸+感染组、感染组和空白组的仔猪结肠内容物样品进行16S rRNA基因高通量测序(Illumina Mi Seq),利用生物信息学软件对各组仔猪肠道微生物菌群多样性、菌群组成及物种差异进行分析。Alpha多样性分析显示,感染组仔猪结肠菌群多样性和丰度最低,菌群失衡;与感染组相比,菊苣酸+感染组仔猪结肠菌群多样性和丰度呈上升趋势。Beta多样性分析显示,菊苣酸+感染组的菌群结构与菊苣酸组、空白组相似,但与感染组存在差异。门、科、属、OUT水平的Venn分析显示,4个实验组中,菊苣酸组仔猪结肠菌群数量最多,感染组仔猪结肠菌群数量最少;菊苣酸+感染组仔猪结肠菌群数量与空白组相近。菌群群落组成分析显示,与空白组、菊苣酸组、菊苣酸+感染组相比,感染组仔猪的厚壁菌门(Firmicutes)、杆菌科(Lactobacillaceae)和乳杆菌属(Lactobacillus)的丰度呈升高趋势,拟杆菌门(Bacteroidota)的丰度降低趋势。菌群物种差异分析显示,互养菌门(Synergistota)的细菌丰度在空白组和菊苣组中呈显著增加,瘤胃球菌种(Ruminococcaceae)的细菌在感染组中的丰度最低。以上结果表明,仔猪口服菊苣酸后再以PDCoV感染,可改善PDCoV感染引起仔猪肠道菌群的失衡,恢复其肠道菌群的多样性和丰度。本研究为菊苣酸改善PDCoV感染引起仔猪肠道菌群的失衡提供了参考依据。To evaluate the effect of cichoric acid on gut microbiota imbalance in piglets infected with porcine deltacoronavirus(PDCoV).In this study,colon content samples from cichoric acid group,cichoric acid+challenge group,challenge group and blank group were selected for 16S rRNA gene high-throughput sequencing(Illumina MiSeq),and the diversity of gut microbiota and composition of bacterial flora in each group were analyzed using bioinformatics software.Alpha diversity analysis showed that the diversity and abundance of colonic flora in the infected group were the lowest,and the flora was unbalanced.Compared with the infected group,the diversity and abundance of colonic flora in the chicory+infected group increased.Beta diversity analysis showed that the colony structure of the cichoric acid+challenge group was similar to cichoric acid group and blank group,but different from that of the infected group.Venn analysis at phylum,family,genus and OUT level showed that among the four experimental groups,the colonic flora in cichoric acid group had the highest abundance,the challenge group had the lowest colonic flora,and the colonic flora in cichoric acid+challenge group was similar to that of the blank group.The analysis of the bacterial community composition showed that the abundance of Firmicutes,Lactobacillaceae and Lactobacillus in challenge group increased compared with blank group,cichoric acid group and cichoric acid+challenge group,while the abundance of Bacteroidota decreased.The bacterial species difference analysis showed that the bacterial abundance of Synergistota was significantly increased in the blank group and chicoric acid group,and the bacterial abundance of Ruminococcaceae was the lowest in the infected group.These results suggest that oral administration of chicoric acid followed by PDCoV reinfection in piglets can improve the imbalance of gut microbiota caused by PDCoV infection and restore the diversity and richness of intestinal flora.This study provides a theoretical basis for the improvement of gu
