摘 要:为了探究柔嫩艾美耳球虫热休克蛋白90(EtHSP90)和组蛋白(EtH4)在体外对鸡胚成纤维细胞(DF-1细胞)凋亡的影响,本试验构建真核荧光表达质粒pEGFP-N1-EtHSP90和pEGFP-N1-EtH4并转染至DF-1细胞,用荧光显微镜观察转染效果,Western blot验证蛋白表达,流式细胞术检测DF-1细胞凋亡。结果显示,经验证成功构建重组质粒pEGFP-N1-EtHSP90和pEGFP-N1-EtH4。流式细胞术检测发现,pEGFP-N1-EtHSP90组早期凋亡率显著低于空白组、pEGFP-N1组和pEGFP-N1-EtH4组(P<0.05);pEGFP-N1-EtH4组早期凋亡率与空白组和pEGFP-N1组相比差异不显著(P>0.05)。因此,EtHSP90早期可抑制细胞凋亡,EtH4既不促进细胞凋亡,也不抑制细胞凋亡。本试验结果为后续深入探究2个蛋白的凋亡机制提供理论基础。To investigate the effects of Eimeria tenella heat shock protein 90(EtHSP90) and histone 4(EtH4) on apoptosis in chicken embryo fibroblast cells(DF-1 cells) in vitro,this study constructed eukaryotic fluorescent expression plasmids pEGFP-N1-EtHSP90 and pEGFP-N1-EtH4 and transfected them into DF-1 cells.The transfection efficiency was observed using a fluorescence microscope,protein expression was verified by Western blot,and apoptosis in DF-1 cells was detected by flow cytometry.The results confirmed the successful construction of recombinant plasmids pEGFP-N1-EtHSP90 and pEGFP-N1-EtH4.Flow cytometry revealed that the early apoptosis rate in the pEGFP-N1-EtHSP90 group was significantly lower than that in the Blank group,pEGFP-N1 group,and pEGFP-N1-EtH4 group(P<0.05).The early apoptosis rate in the pEGFP-N1-EtH4 group did not differ significantly from the Blank group and the pEGFP-N1 group(P>0.05).Therefore,EtHSP90 can inhibit early cell apoptosis,while EtH4 neither promotes nor inhibits cell apoptosis.These results provide a theoretical basis for further exploration of the apoptosis mechanisms of these two proteins.
