摘 要:为建立一种黔产多须公Eupatorium chinense Linn.的薄层色谱鉴别方法,参照薄层色谱法(通则0502),以槲皮素为标准品,对展开剂、点样量、提取方式、提取溶剂、显色剂、检视波长、不同厂家硅胶G板、环境条件(湿度、温度)等因素进行了方法学考察.结果显示,多须公药材薄层色谱鉴别最佳方法为供试品点样量为5μL、标准品点样量为2μL、展开剂为甲苯-乙酸乙酯-甲酸(5∶2∶1)、显色剂为3%三氯化铝乙醇溶液、显色温度为105℃、检视波长为365 nm,此外,研究发现,提取溶剂、提取方式和不同厂家硅胶G板及环境条件(湿度、温度)对薄层色谱法的鉴别影响不大.本方法操作简单,专属性强,稳定性及重现性良好,可用于黔产多须公药材薄层色谱定性鉴别.In order to establish a thin-layer chromatography(TLC)identification method for Eupatorium chinense Linn.produced in Guizhou province,a methodological study was conducted using Quercetin as the standard substance,following the TLC method(General Rule 0502).Factors such as developing agent,sample size,extraction method,extraction solvent,chromogenic agent,inspection wavelength,silica gel G plates from different manufacturers,and environmental conditions(humidity and temperature)were investigated.The results showed that the best method for TLC identification of E.chinense medicinal herb is:the sample size of the test sample is 5μL.The standard sample size is 2μL.The developing agent is toluene ethyl acetate formic acid(5∶2∶1),the color developing agent is aluminum chloride ethanol solution of 3%,the color developing temperature is 105℃,and the inspection wavelength is 365 nm.In addition,research has found that the extraction solvent,extraction method,silica gel G plates of different manufacturers,and environmental conditions(humidity and temperature)have little effect on the identification of TLC.Overall,this method is simple to operate,has strong specificity,good stability and reproducibility,and can be used for the qualitative identification of Guizhou produced Eupatorium chinense Linn by TLC.
