摘  要:目的基于网络药理学方法筛选益气甘露方的有效成分、作用靶点及信号转导通路,探讨益气甘露方治疗糖尿病的潜在作用机制。方法筛选益气甘露方潜在活性成分及其对应靶点,应用Cytoscape 3.9.1软件对成分和靶点进行网络构建与分析。在GeneCards、TTD等数据库中筛选糖尿病相关靶点,取药物有效成分靶点与疾病靶点交集,在Metascape数据库中进行基因注释的GO分析和信号通路的KEGG富集分析。结果通过筛选得到益气甘露方活性成分(口服生物利用度(OB)≥30%且类药性(DL)≥0.18)157项,药物与疾病共同靶点247项,有效成分主要为槲皮素、山柰酚、7-O-甲基异木豆醇等,主要调节的通路为脂质与动脉粥样硬化信号通路,除此之外,还有PI3K-Akt信号通路、MAPK信号通路等。结论益气甘露方可能是通过调控脂质与动脉粥样硬化信号通路、PI3K-Akt信号通路、MAPK信号通路等调节血糖代谢。Objective To explore the effective ingredients,targets,signal transduction pathways and potential mechanism of Yiqi Ganlu Formula in the treatment of diabetes by using the network pharmacology.Method The potential active ingredients and corresponding targets were obtained.The component and target network was constructed and analyzed with Cytoscape 3.9.1 software.The related targets of diabetes were screened in Gene Cards,TTD and other databases.The targets obtained through drug screening and disease screening were inter-sected,GO analysis of gene annotation and KEGG enrichment analysis of signal pathways were conducted in Met-ascape database.Results 157 active ingredients(oral bioavailability(OB)≥30%and drug like(DL)≥0.18)and 247 common drug-disease targets of Yiqi Ganlu Formula were obtained.The active ingredients were quer-cetin,kaempferol,7-O-methylisomucronulatol,etc.The main regulatory pathways were lipid and atherosclerotic pathway,PI3K-Akt signaling pathway,MAPK signaling pathway,etc.Conclusion Yiqi Ganlu Formulas may affect the blood glucose metabolism by regulating lipid and atherosclerotic pathway,PI3K-Akt signaling pathway,MAPK signaling pathway,etc.
