摘 要:目的探究可替代巴比妥缓冲液的新型血清蛋白电泳缓冲液。方法通过不同电压、电泳时间梯度的电泳,筛选新型电泳缓冲液的最佳试验条件;比较新型电泳缓冲液与巴比妥缓冲液分离血清蛋白的图谱效果、蛋白区带分散程度及各组蛋白质相对含量。结果新型电泳缓冲液在电压为70V、电泳时间为50min时,电泳图谱稳定且重复性更好,其图谱出现5条区带的比率比巴比妥缓冲液高23.14%、蛋白区带分散程度适中。新型缓冲液膜条扩展的总距离、最远区带与最近区带的距离均短于巴比妥缓冲液,差异有统计学意义(P<0.05)。新型缓冲液与巴比妥缓冲液点样区距第一条带远端的距离比较差异无统计学意义。两种缓冲液分离的各组蛋白相对含量比较差异无统计学意义。结论新型电泳缓冲液分离血清蛋白图谱区带清晰、重复性好、结果稳定,电泳图谱效果、蛋白区带分散程度优于巴比妥缓冲液,且在电压为70V、电泳时间为50min试验条件下,与巴比妥缓冲液分离的各种蛋白含量差异不显著,可替代其用于血清蛋白的醋酸纤维素薄膜电泳实验中。Objective To investigate the new serum protein electrophoresis buffer that can replace barbiturate buffer.Methods The optimum test conditions for the new electrophoresis buffer were screened by electrophoresis at different voltages and electrophoresis time gradients;and the graphical results,the degree of dispersion of the protein bands and the relative content of each protein were compared between the new electrophore-sis buffer and barbiturate buffer for the separation of serum proteins.Results The results showed that the new electrophoresis buffer was stable and reproducible at a voltage of 70 V and an electrophoresis time of 50 min,the rate of five bands was 23.14%,which was higher than that of the barbitu-rate buffer,and the protein zone dispersion was moderate.The total distance and the distance between the furthest zone and the nearest zone of the new buffer were shorter than those of the barbital buffer,and the differences were statistically significant(P<0.05).There was no significant differ-ence in the distance between the new buffer and the barbiturate buffer from the far end of the first band.There was no significant difference in the rel-ative protein content between the two buffers.Conclusion The new electrophoretic buffer isolated serum protein map with clear bands,good repeat-ability and stable results.The electrophoretic map effect and protein band dispersion degree were better than that of barbiturate buffer.Under the test conditions of 70 V and 50 min electrophoresis time,there was no significant difference in the content of various proteins separated from barbiturate buffer,it can be substituted for cellulose acetate film electrophoresis of serum protein.
