摘 要:目的探讨芹菜素对实验性代谢综合征(MS)的干预效果。方法采用高脂肪/高果糖饮食诱导大鼠MS模型,随机分为正常组、模型组、芹菜素低剂量组(25mg/kg)、芹菜素中剂量组(50mg/kg)、芹菜素高剂量组(100mg/kg)、阳性对照组(二甲双胍50mg/kg);灌胃给药8周,称体质量、内脏脂肪质量,计算脂体比;检测空腹血糖,取血清检测总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)、游离脂肪酸(FFA)及胰岛素水平,计算胰岛素抵抗模型评估指数(HOMA-IR)。结果与模型组相比,芹菜素各剂量组可明显降低MS大鼠的体质量、内脏脂肪质量及脂体比(P<0.01),显著下调空腹血糖及血清TC、TG、LDL-C、FFA水平(P<0.05或P<0.01),高剂量组可显著升高HDL-C水平(P<0.05),高、中剂量组可显著降低胰岛素水平和HOMA-IR指数(P<0.05或P<0.01)。结论芹菜素通过减少腹部脂肪堆积、纠正糖脂代谢紊乱、缓解胰岛素抵抗状态等途径,干预实验性MS的病变进程。Objective To explore the treatment effect of apigenin on rats with high fat/high fructose diet-induced metabolic syndrome(HFD-MS).Methods HFD-MS model in rats was induced by a 16 weeks of high-fat/high-fructose diet.Rats were randomly divided into normal,model,apigenin low,medium and high doses(25,50 and 100mg/kg,respectively),and positive control(metformin 50mg/kg)groups.The drugs were administered by gavage for 8 weeks.The body and visceral fat weights were measured,and the fat-body ratio was calculated.The blood glucose,total cholesterol(TC),triglyceride(TG),low-density lipoprotein(LDL-C),high-density lipoprotein(HDL-C),free fatty acid(FFA),and insulin levels in serum were detected,and the homeostasis model assessment of insulin resistance(HOMA-IR)was calculated.Results Compared with the model group,apigenin administration significantly reduced the body and visceral fat weights and fat-body ratio in HFD-MS rats(P<0.01),significantly down-regulated fasting blood glucose and serum TC,TG,LDL-C,and FFA levels(P<0.05 or P<0.01),and the high-dose of apigenin evidently increased HDL-C levels(P<0.05),while the high and medium doses of apigenin significantly reduced insulin levels and HOMA-IR(P<0.05 or P<0.01).Conclusion Apigenin intervenes the pathological process of HFD-MS by reducing the accumulation of abdominal fat,correcting the disorders of glucose-lipid metabolism,and alleviating the state of insulin resistance.
