摘  要:目的 比较两种脱水程序和5种浸蜡时间对小鼠肝、肠道组织石蜡切片质量的影响。方法 以BALB/c小鼠肝和肠道组织为材料,按照两种不同的脱水程序(程序一、程序二)和5种不同的浸蜡程序(浸蜡总时间为90、120、150、180和210 min)制备石蜡切片,比较其组织切片和HE染色的效果。结果 脱水程序一制作的石蜡切片容易碎裂,皱褶明显,组织中央易见空洞;脱水程序二制作的石蜡切片完整无空洞,石蜡切片经HE染色后镜下观察结构完整清晰。相较于其他4种浸蜡程序,浸蜡总时间为150 min时制作的石蜡切片染色后观察最完整清晰。结论 使用脱水程序二及浸蜡总时间为150 min时所制作的石蜡切片质量有所提升,易于镜下观察,推荐作为小鼠肝和肠道组织石蜡切片的优化程序。Objective To compare the effects of two dehydration procedures and five wax immersion times on the quality of paraffin sections of mouse liver and intestinal tissues,with a view to optimizing the preparation procedures of paraffin sections.Method The liver and intestinal tissues of BALB/c mice were prepared in two different dehydration procedures(Procedure I and Procedure II)and five different wax immersion procedures(total immersion time of 90,120,150,180 and 210 min)to compare the result of tissue sectioning and HE staining.Result The paraffin sections produced by dehydration procedure 1 were easily fragmented,with obvious wrinkles and cavities in the center of the tissue;the paraffin sections produced by dehydration procedure 2 were intact and free of cavities,and the structure of the paraffin sections was observed to be intact and clear under the microscope after HE staining.Compared with the other four wax immersion procedures,the paraffin sections produced with a total immersion time of 150 min were the most complete and clear after staining.Conclusion The quality of the paraffin sections produced by using the dehydration procedure II and the total time of wax immersion of 150 min was improved and easy to observe under the microscope,which is recommended as the optimized procedure for paraffin sections of mouse liver and intestinal tissues.
