药用大黄内生真菌的分离及鉴定

摘　　要：药用大黄可以加强胃肠道蠕动、清热解毒、止血、保肝利胆、改善肾功能、抗氧化、抗血栓、抗炎抑菌、抗肿瘤等。而内生真菌则是植物体内广泛存在的一种重要微生物资源,部分内生真菌对植物生长发育具有重要影响。大黄内生真菌与大黄的生长、药用成分的生成等之间的关系鲜有报道,这些研究都以内生真菌的分离鉴定为前提。因此,本实验以药用大黄根部为材料,经过严格的表面消毒,分离纯化得到内生真菌,观察内生真菌的菌落培养形态、个体显微特征,并根据ITS序列进行分类鉴定。结果表明:综合形态学观察和分子生物学鉴定实验结果,共分离出26株内生真菌,13个种,分属于8个属,其中2株真菌的分子鉴定结果相似度系数较低,值得进一步研究。该结果对研究大黄内生真菌和大黄的生长关系以及大黄药用成分的关系奠定了基础。Rheum Officinale Baill is a genus of rheum,which is of great significance in medicinal value.However,endophytes are an important microbial resource widely found in plants,and some endophytes have an important influence on plant growth and development.The growth relationship between Rheum Officinale Baill endophytes and Rheum Officinale Baill and the relationship between medicinal components of Rheum Officinale Baill are rarely reported,and these studies are based on the isolation and identification of endogenous fungi.Therefore,we conducted relevant studies.After using Rheum Officinale Baill root,endophytic fungi were isolated and purified through strict surface disinfection,and the colony culture morphology and individual microscopic characteristics of endophytic fungi were observed,and classified according to the ITS sequence.Based on the morphological observation and molecular biology identification experiments,26 endophytic fungi and 13 species were isolated into 8 genera,of which two strains showed low similarity,which deserves further study.The results lay a foundation for studying the growth relationship between Rheum Officinale Endophytes and Rheum Officinale and the medicinal ingredients of Rheum Officinale..
