摘  要:肌动蛋白相关蛋白(Actin-related proteins,ARPs)是一类和肌动蛋白同源的蛋白,其中核定位的ARP基因主要是以染色质重塑复合体成员被研究,染色质重塑复合体成员参与基因的表达调控进而影响生物体的生长发育过程。为揭示ARPs在谷子中的功能,将拟南芥中核定位的5个成员在谷子中进行同源比对,获得了5个谷子ARP基因,分别是SiARP4、SiARP5、SiARP6、SiARP7、SiARP9。然后对这5个成员的基因结构、理化性质、蛋白保守基序等信息进行分析预测,同时利用实时荧光定量PCR(RT-qPCR)对逆境胁迫下SiARP4和SiARP7表达情况进行了检测。结果表明,这5个成员均为亲水性蛋白质,保守基序的组成存在一定差异,都含有Actin-like ATPase结构域。系统进化分析显示,谷子ARPs与玉米和高粱ARP蛋白的亲缘关系较近。启动子元件分析结果显示,上述5个基因的启动子含有光响应、激素响应、应激响应及其他类生长调控元件。RT-qPCR分析表明,ARP4和ARP7基因在干旱胁迫和盐胁迫下发生了不同程度的表达量变化,说明ARP4和ARP7基因参与到逆境胁迫响应过程中,研究为染色质重塑组分ARPs参与谷子抗逆过程的分子机制解析奠定了理论基础。The actin-related proteins(ARPs)are homologous to the conventional actin,and nucleus ARP are regarded as components of chromatin remodeling complex.Chromatin remodeling complexes play important roles in regulation of gene transcriptional expression,and involve in the development processes in plant.In order to reveal the function of ARPs in foxtail millet,the ARP members in foxtail millet were identified by sequence alignment with the five nuclear localization genes in Arabidopsis,which are SiARP4,SiARP5,SiARP6,SiARP7 and SiARP9.Then gene structure,physicochemical properties,conserved protein motifs and other traits were analyzed and predicted.The real-time quantitative PCR(RT-qPCR)was used to detect the expression level of SiARP4 and SiARP7 under different stresses.The results showed that the five members were hydrophilic protein and there were some difference in the composition of the conserved motifs,but all contained Actin-like ATPase domain.The phylogenetic tree revealed that SiARP protein had a more closely relationship with ARP proteins in maize and sorghum.The promoters of ARPs in foxtail millet contain light response element,hormone response,stress response and other regulatory elements.The result of RT-qPCR showed that expression levels of ARP4 and ARP7 changed in different degrees under drought stress and salt stress,indicating that ARP4 and ARP7 involved in stress response.This study provided theoretical support for further analysis on responding mechanism of chromatin remodeling component ARPs to stress resistance of millet.
