摘  要:为建立一种可以简单快速区分复合四倍体鲫与其双亲的方法,根据鲫和鲤生长相关基因的差异,进行了DNA分子标记的开发,最终筛选得到一对引物,利用该分子标记在兴国红鲤和异育银鲫“中科3号”基因组中分别扩增出1 399 bp和1 125 bp的DNA片段,并且在复合四倍体鲫中同时扩增出这两个DNA片段。试验结果显示,这种分子标记提供了一种能较为快速、高效区分亲本兴国红鲤和异育银鲫“中科3号”及其子代复合四倍体鲫的方法,可为今后育种研究中苗种或成鱼的鉴定提供帮助。In order to establish a simple and rapid method to distinguish between multiple tetraploid allogynogenetic crucian carp and its parentage,DNA molecular markers were developed and a pair of primers was screened based on the difference of growth-related genes between Carassius auratus and Cyprinus carpio sequences.The DNA fragments of 1399 bp and 1125 bp were amplified in Xingguo red carp(C.carpio var.singuonensis)and C.auratus gibelio“CASⅢ”,respectively.Moreover,both 1399 bp and 1125 bp were amplified in multiple tetraploid allogynogenetic crucian carp.In summary,this molecular marker provides a relatively fast and efficient method for distinguishing between C.carpio var.singuonensis,C.auratus gibelio“CASⅢ”and multiple tetraploid allogynogenetic crucian carp,and also provides assistance for the identification of fry or adult fish in future breeding research.
