猪附红细胞体感染家兔模型的建立

摘　　要：为了建立猪附红细胞体感染家兔模型。将摘除脾脏后的12只家兔随机平均分为3组。A组为模型组,接种2 mL纯化的猪附红细胞体;B组为阴性对照组,接种2 mL猪附红细胞体阴性血液分离物;C组为空白对照组,接种2 mL生理盐水。接种后通过测量体温、体质量、PCR检测、血常规、血液生化指标和病理组织学检验来鉴定猪附红细胞体感染情况。结果显示,A组家兔与B组、C组相比,体温、体质量变化差异显著(P<0.05)。A组家兔出现了消瘦、被毛杂乱、结膜黄染等临床症状。血常规检测结果显示,A组家兔红细胞总数、平均体积、血红蛋白含量均减少,差异显著(P<0.05);血红蛋白、血红蛋白浓度均减少,差异极显著(P<0.01),提示家兔贫血。血液生化指标检测结果显示,A组家兔尿素、白球比、尿酐比,丙氨酸氨基转移酶均升高,差异显著(P<0.05);球蛋白、肌酐、血糖均下降,差异显著(P<0.05),提示家兔免疫力下降,肝功能和肾功能受损。病理组织学观察结果显示,A组家兔肝脏充血、水肿、肺脏局部有炎性细胞、心肌细胞出血。试验结果表明本研究成功建立了猪附红细胞体感染家兔模型。To develop a rabbit model of Mycoplasma suis infection,12 rabbits were performed for spleen removal and randomly divided into three groups.Group A was the model group and received 2 mL purified M.suis.;Group B served as negative control and received2 mL porcine M.suis negative blood sample.Group C was used as blank control and administered with 2 mL normal saline.After inoculation,rabbits were observed for M.suis infection by body temperature and weight measurements,PCR detection,blood routine and biochemical index checks and histopathological examination.The results showed that the body temperature and weight of rabbits in group A were significantly different from those in group B and group C(P<0.05).Rabbits in group A appeared thin,fur clutter,conjunctival yellow stains and other clinical signs.The results of blood routine testing showed that total number of red blood cells,average volume and hemoglobin content of rabbits in group A significantly decreased(P<0.05).The concentrations of hemoglobin and hemoglobin significantly decreased(P<0.01),suggesting anemia occurred in rabbits.The results of blood biochemical indexes showed that urea,white ball ratio,urinary anhydride ratio and alanine aminotransferase of rabbits in group A significantly increased(P<0.05).Globulin,creatinine and blood glucose also significantly decreased(P<0.05),suggesting that the decreased immunity and damaged liver and renal functions.The histopathological observations on rabbits in group A showed hyperemia and edema in livers and infiltration of inflammatory cells and myocardial cell hemorrhage in lungs.These results showed that a rabbit model of M.suis infection was successfully developed in this study.
