摘  要:苹果酸脱氢酶(malate dehydrogenase,MDH)在植物生长发育过程中发挥着至关重要的作用,然而在植物免疫中的作用,仍然是难以捉摸的。本研究在木薯中发现了一个MeMDH1基因,以MeMDH1为诱饵蛋白构建载体p GADT7-Me MDH1,从木薯cDNA文库筛选互作蛋白。得到一个候选基因MeLSD3,研究报道LSD在细胞程序性死亡以及植物光合作用和生长等生物过程中发挥重要作用。我们通过酵母双杂交(Yeast two-hybrid,Y2H)和双分子荧光互补(bimolecular fluorescence complementation,BiFC)实验验证了MeLSD3和MeMDH1在木薯体内体外的互作。为进一步探究木薯苹果酸脱氢酶在植物抗病反应中的功能及其调控的分子机制提供理论基础。Malate dehydrogenase(MDH)plays a crucial role in plant growth and development,yet its role in plant immunity remains elusive.In this study,a MeMDH1 gene was identified in cassava,and the vector pGADT7-MeMDH1 was constructed using MeMDH1 as a decoy protein to screen the reciprocal proteins from a cassava cDNA library.A candidate gene MeLSD3,was obtained and studies reported that LSD plays an important role in biological processes such as programmed cell death and plant photosynthesis and growth.We verified the interaction between MeLSD3 and MeMDH1 in cassava in vitro and in vivo by yeast two-hybrid(Y2H)and bimolecular fluorescence complementation(BiFC)experiments.It provides a theoretical basis for further investigation of the function of cassava malate dehydrogenase in plant disease resistance response and the molecular mechanism of its regulation.
