摘  要:葛拟锈病是近年来制约葛根产业发展的重大病害,导致产区种植葛品种产量和品质逐年下降,给种植户造成较大的经济损失。葛拟锈病是由葛藤集壶菌侵染引起的真菌病害,而防御性酶和丙二醛(MDA)是反映寄主防卫反应能力和抗性的重要生化指标。本研究以课题组选育的感病品种‘桂葛8号’和抗病品种‘桂葛18号’为供试材料,采用人工活体接种的方法,研究葛藤集壶菌侵染后不同抗性品种超氧歧化物酶(SOD)、多酚氧化酶(PPO)、过氧化氢酶(CAT)和几丁质酶(CHI)的活性及丙二醛(MDA)含量的变化。结果表明,葛藤集壶菌侵染后两个品种的CAT、PPO、SOD及CHI酶活性和MDA含量变化显著。两个品种间,不论与对照组还是与处理组相比,抗病品种‘桂葛18号’的CAT、PPO、SOD及CHI酶活性均显著高于感病品种‘桂葛8号’,增幅更大,并且在较高活性水平持续更长时间。而MDA含量则刚好相反,感病品种‘桂葛8号’显著高于抗病品种‘桂葛18号’,增幅更大,并且在较高活性水平持续更长时间。两个品种自身的防御性酶活性和MDA含量存在显著差异,说明防御酶与葛根抗病性有一定的相关性,其应对病原菌侵染的响应快慢差异反映了葛根品种抗拟锈病菌的程度。相较敏感型品种,抗性品种对葛藤集壶菌侵染的响应速度更快,抗性活力更高。Pueraria pseudo-rust is a serious disease that restricts the development of pueraria industry in recent years,which leads to the decrease of the yield and quality of pueraria varieties year by year,and causes great economic losses to growers.Pueraria pseudo-rust is a fungal disease caused by Synchytrium puerariae Miy.Defensive enzyme and malondialdehyde(MDA)are important biochemical indexes of host defense response and resistance ability.The sensitive cultivar'Guige8'and resistant cultivar'Guige18'which selected by our research group were used as test materials to study the activities of superoxide dismutase(SOD),polyphenol oxidase(PPO),catalase(CAT)and chitinase(CHI),as well as malondialdehyde(MDA)content changes by artificial living inoculation.The results showed that the activities of four defense enzymes and MDA content of the two pueraria varieties changed significantly after being infected by Synchytrium puerariae Miy.The CAT,PPO,SOD and CHI defense enzyme activities of the resistant cultivar'Guige18'were significantly higher than those of the sensitive cultivar'Guige8'in both the control group and the treatment group,with a greater increase rate and a longer duration at a higher activity level.On the contrary,the MDA content of'Guige8'was higher than that of'Guige18'significantly and maintained at a higher level for a longer time.There were significant differences in the defense enzymes activities and MDA contents between the two varieties of pueraria.The results showed that there was a certain correlation between defense enzyme and disease resistance of pueraria.The difference of response speed to pathogen infection reflected the degree of resistance of pueraria varieties to Synchytrium puerariae Miy.Compared with the sensitive cultivar,the resistant one had faster response speed and higher resistance activity to Synchytrium puerariae Miy infection.
