摘 要:半胱氨酸(Cys)在生物体系中起着不可缺少的作用,可以参与人体内的许多生理和病理过程,对氧化还原稳态和细胞活性的调节有重要作用。大量研究表明,Cys浓度异常与许多疾病的发生密不可分,因此有必要开发特异性检测Cys的荧光探针。本文以丙烯酰基为识别单元,以二氰基异佛尔酮为荧光基团,设计合成了一种荧光探针ISO-EV-AC。实验结果表明,探针ISO-EVAC对Cys的检测展现出较高的选择性,不受其他分析物的干扰,检测限低至31 nmol/L,响应Cys的时间为5 min。细胞实验结果表明探针ISO-EV-AC可用于检测细胞中的Cys。Cysteine(Cys)plays an indispensable role in biological systems,which can participate in a variety of physiological and pathological processes in the human body and plays an essential role in the regulation of redox homeostasis and cell activity.A large number of studies have shown that abnormal Cys concentrations are strongly associated with the occurrence of various diseases.Therefore,it is necessary to develop a specific fluorescence probe for Cys detection.In this paper,a fluorescent probe ISO-EV-AC was designed and synthesized using acryloyl group as the recognition unit and isophorone dicyano as the fluorophore.The results indicated that probe ISO-EV-AC exhibits a high selectivity for the detection of Cys without interference from the other nanlytes.The detection limit was as low as 31 nmol/L and the response time of Cys was 5 min.The results of the cell experiments demonstrated that probe ISO-EV-AC can be used to detect Cys in cells.
