摘 要:本文以染料的脱色率作为主要的考察指标,利用红芝LYL263与苏平平菇共培养所产的漆酶粗酶液,对甲基橙、活性艳蓝、孔雀绿、溴酚蓝和结晶紫等5种染料进行脱色降解研究,考察了pH、反应温度、反应时间和漆酶酶活等条件对脱色降解效果的影响。结果表明,红芝LYL263与苏平平菇共培养所产的漆酶粗酶液,对这5种染料均具有较好的脱色降解效果,对脱色降解条件进行优化后,这些染料的脱色率得到显著提升。漆酶粗酶液中含有的低分子物质可能起到了介体的作用,促进了漆酶对非底物型染料(甲基橙、孔雀绿、溴酚蓝和结晶紫)的脱色降解。当脱色反应的pH=4、反应温度为40℃、漆酶酶活为100U·mL^(-1)时,甲基橙和活性艳蓝的脱色率最高分别达到73.2%和96.5%;当pH=4、反应温度为30℃、漆酶酶活为100 U·mL^(-1)时,孔雀绿的脱色率最高达到96.6%;当pH=3、反应温度为40℃、漆酶酶活为100U·mL^(-1)时,溴酚蓝的脱色率最高达到77.5%;当pH=4、反应温度为30℃、漆酶酶活为75U·mL^(-1),结晶紫的脱色率最高达到88.5%。With the decolorization rate of dyes as the main index,the decolorization and degradation of five dyes,included methyl orange,active brilliant blue,malachite green,bromophenol blue and crystal violet,were studied using the crude laccase enzyme produced by the co-culture of LYL263 and Pleurotus ostreatus.The effects of pH,reaction temperature,reaction time and laccase activity on the decolorization and degradation were investigated.The results showed that the crude laccase produced from the coculture of LYL263 and Pleurotus ostreatus had good decolorization and degradation effect on these five dyes,and the decolorization rate of these dyes was significantly improved after the optimization of decolorization and degradation conditions.When pH=4,reaction temperature was 40℃and laccase activity was 100 U/mL,the decolorization rates of methyl orange and reactive brilliant blue reached 73.2%and 96.5%,respectively.At pH=4,reaction temperature 30℃and laccase activity 100 U/mL,the decolorization rate of malachite green reached 96.6%.At pH=3,reaction temperature 40℃and laccase activity 100 U/mL,the decolorization rate of bromophenol blue reached 77.5%.At pH=4,reaction temperature 30℃and laccase activity 75 U/mL,the decolorization rate of crystal violet reached 88.5%.
