摘 要:目的对桑树、柳树、油茶树、夹竹桃树、苦楝树和漆树6种寄主植物的桑寄生水提物进行斑马鱼胚胎发育毒性研究,探讨寄主植物对桑寄生药材胚胎毒性影响,为桑寄生安全用药提供理论参考。方法以受精且发育正常6 hpf的斑马鱼胚胎为胚胎模型,采用桑树、柳树、油茶树、夹竹桃树、苦楝树和漆树6种寄主植物的桑寄生水提物梯度浓度处理斑马鱼胚胎,统计72 hpf斑马鱼胚胎的死亡率,采用Graphpad Prism 8.0软件绘制量-毒曲线,计算半数致死浓度(LC_(50))和10%致死浓度(LC_(10))。按低(1/2LC_(10))、中(LC_(0))、高(LC_(10))3个药物浓度组处理6 hpf斑马鱼胚胎,记录24 hpf斑马鱼胚胎在1 min内的自主运动次数,及72 hpf斑马鱼胚胎的孵化率、眼睛大小及体长。结果桑树、柳树、油茶树、夹竹桃树、苦楝树和漆树寄主桑寄生水提物LC50分别为:6.28、3.86、2.14、1.02、1.00、0.36 mg/mL,LC_(10)分别为:4.77、2.76、1.16、0.67、0.62、0.25 mg/mL。与空白组相比,桑树寄主桑寄生水提物低、中、高药物浓度组斑马鱼的胚胎自主运动次数、孵化率和仔鱼眼睛大小、生长体长均没有明显变化(P>0.05);其它5种寄主桑寄生水提物的中、高药物浓度组的斑马鱼胚胎自主运动次数明显减少,孵化率降低,仔鱼的眼睛出现明显或非常明显变小,体长明显或非常明显变短(P<0.05,P<0.01)。结论除桑树寄主桑寄生水提物没有观察到明显胚胎发育毒性外,其它5种寄主桑寄生水提物对斑马鱼胚胎发育均表现出不同程度的毒性。Objective To study the embryonic toxicity of the aqueous extracts of Taxilli Herba from Morus alba,Salix babylonica,Camellia oleifera,Nerium indicum,Melia azedarach and Toxicodendron trichocarpum to zebrafish embryos,hoping to provide a theoretical basis for the safe use of Taxilli Herba.Methods The normally developing zebrafish embryos at 6 hours post-fertilisation(hpf)was used as an animal model.Zebrafish embryos were treated with gradient concentrations of the aqueous extracts of Taxilli Herba from Morus alba,Salix babylonica,Camellia oleifer,Nerium indicum,Melia azedarach and Toxicodendron trichocarpum,and zebrafish embryo mortality was counted at 72 hpf.GraphPad Prism 8.0 was used to plot the dose-toxicity curve after calculating the median lethal concentration(LC_(50))and the 10%lethal concentration(LC_(10)).Three concentration groups of low-,medium-,and high-dose were established according to their 1/2LC_(10),LC_(0),LC_(10),and treated to zebrafish embryos at 6 hpf.Then,the number of voluntary movements within 1 minute was then counted at 24 hpf.Hatching rate,the eye size,and body length were counted and measured at 72 hpf.Results The results show that the LC_(50) values of aqueous extracts of Taxilli Herba from six hosts:M.alba,S.babylonica,C.oleifera,N.indicum,M.azedarach,and T.trichocarpum were 6.28,3.86,2.14,1.02,1.00 and 0.36 mg/mL,respectively.The LC_(10) values were 4.77,2.76,1.16,0.67,0.62 and 0.25 mg/mL,respective ly.In comparison to the blank group,no significant alterations were observed in the number of voluntary movements,hatching rate,eye size,and body growth length in all the dose groups of Taxilli Herba extracted from M.alba(P>0.05).In contrast,the medium-and high-dose groups of the aqueous extracts of Taxilli Herba from the other five hosts showed significant changes(P<0.05,P<0.01).Conclusion Taken together,with the exception of Taxilli Herba from M.alba,Taxilli Herba from the other five hosts exhibited varying degrees of embryonic toxicity to zebrafish embryos.
