摘 要:目的探究纳米氧化铈亚急性暴露对小鼠学习记忆能力的影响。方法鼻腔滴注染毒昆明小鼠30 d后进行Morris水迷宫实验,取海马组织进行尼氏染色,测定海马中超氧化物歧化酶(SOD)、丙二醛(MDA)、γ-氨基丁酸(GABA)和谷氨酸(Glu)含量,并采用Western blot检测炎性蛋白NF-κB和NLRP3表达量。结果相对于对照组,中和高染毒组小鼠行为学检测出现学习记忆能力障碍,中和高剂量组尼氏染色阳性细胞数减少,差异有统计学意义(F=9.246,P<0.05);高剂量组SOD活性降低,差异有统计学意义(F=3.969,P<0.05),中和高剂量组MDA含量升高,差异有统计学意义(F=9.368,P<0.05);各剂量组的GABA含量呈降低趋势,差异有统计学意义(F=59.81,P<0.05),中和高剂量组Glu含量升高差异有统计学意义(F=10.64,P<0.05);高剂量组NF-κB、NLRP3表达量增加,差异有统计学差异(F=10.97,F=7.443,P<0.05)。结论本试验条件下,一定剂量的纳米氧化铈亚急性染毒可影响小鼠空间学习能力,损伤海马神经元,影响其抗氧化能力并产生炎症反应。Objective To investigate the effects of sub-acute exposure to cerium oxide nanoparticles on cognitive impairment in mice.Methods Morris water maze test were performed after intranasal instillation 30 days.The hippocampus was removed for Nissl staining and the contents of superoxidedismutase(SOD),malondialdehyde(MDA),γ-aminobutyric acid(GABA)and glutamate(Glu)in hippocampus were determined,the expression of NF-κB and NLRP3 by Western blot.Results Compared with the control group,the middle and high-dose groups had learning impairment in behavioral tests,and the middle and high dose groups had a significant reduction in the number of Nissl staining positive cells(F=9.246,P<0.05).The activity of SOD in the high-dose group decreased,and the difference was statistically significant(F=3.969,P<0.05).The content of MDA in the middle and high-dose groups increased,and the difference was statistically significant(F=9.368,P<0.05).The content of GABA in each dose group decreased,and the differences were statistically significant(F=59.81,P<0.05).The content of Glu in the middle and high dose groups increased,and the differences were statistically significant(F=10.64,P<0.05).The expression of NF-κB and NLRP3 in the high dose group were significantly increased(F=10.97,F=7.443,P<0.05).Conclusion Under the conditions of this test,sub-acute exposure to cerium oxide nanoparticles can affect the spatial learning ability of mice,damage hippocampal neurons,affect their antioxidant capacity and induce inflammatory response.
