摘 要:以总黄酮和总皂苷的综合评分为考察指标,采用Box-Behnken法优化提取工艺最佳的乙醇浓度、提取时间、料液比及浸泡时间。考察提取物的ABTS+及DPPH清除率及对在H 2 O 2诱导的氧化损伤B16细胞存活率,SOD、CAT和MDA水平评价其抗氧化活性。结果显示,最佳提取工艺为:13.45倍量67.86%乙醇溶液浸泡0.89 h,回流1.68 h提取3次。锦灯笼提取物具有良好的抗氧化活性,可提高细胞CAT和SOD活性,降低MDA含量;提高氧化损伤状态的B16细胞的存活率,为深入研究锦灯笼提供实验基础。Using the comprehensive scores of total flavonoids and total saponins as the evaluation indicators,the Box Behnken method was used to optimize the extraction process for the optimal ethanol concentration,extraction time,solid-liquid ratio and soaking time.The ABTS+and DPPH clearance rates of the extract were assessed,as well as its antioxidant activity against H 2 O 2 induced oxidative damage to B16 cells and the levels of SOD,CAT,and MDA.The results showed that the optimal extraction process was as follows:13.45 times the amount of 67.86%ethanol solution soaked for 0.89 h,refluxed for 1.68 h,and extracted three times.Jindeng Lantern extract had good antioxidant activity,which can increase cellular CAT and SOD activities and reduce MDA content.Improving the survival rate of B16 cells in oxidative damage state provided an experimental basis for in-depth research on Jindeng lanterns.
