双参瓜蒌汤对肺动脉高压模型大鼠血管活性物质的影响

摘　　要：目的观察双参瓜蒌汤对肺动脉高压模型大鼠肺部血管紧张素及血清炎性因子的影响。方法将50只大鼠随机平均分为5组:空白对照组、模型对照组、中药高剂量组、中药低剂量组、西药组(依那普利)。后4组腹腔一次性注射70 mg/kg野百合碱,诱导30 d后,形成肺动脉高压模型。中药高剂量组、中药低剂量组每日中药灌胃,5 mL/d。西药组采用依那普利(0.05 mg/d)灌胃。空白对照组、模型对照组给予5 mL氯化钠溶液灌胃。干预4周后,采用酶联免疫吸附试验(ELISA)检测肺组织内血管紧张素转换酶2(ACE2)、血管紧张素1-7(Ang1-7),血清中炎性因子:磷脂酶A2(PLA2)、血清淀粉样蛋白A(SAA)水平。结果与模型组比较,中药高剂量组、中药低剂量组、西药依那普利组ACE2、Ang-1-7水平均有不同程度降低,其中中药高剂量组降低最为明显,依那普利组略有降低。与模型组比较,中药高剂量组、中药低剂量组炎性标志物PLA2、SAA水平均有不同程度降低,其中中药高剂量组降低最为明显。结论双参瓜蒌汤具有下调ACE2、Ang1-7水平,起到扩张肺部血管的作用,同时下调炎性因子PLA2、SAA水平,减少血管内皮细胞炎症性凋亡,延缓血管内壁重塑,这可能是中药治疗肺动脉高压作用机制之一。Objective To observe the effects of ShuangShen Gualou Decoction(双参瓜蒌汤)on pulmonary angiotensin and vascular endothelial cell inflammatory factors in rats with pulmonary hypertension.Methods 50 rats were randomly divided into 5 groups:blank control group,model control group,high dose group,low dose group and western medicine enalapril group.After 30 days of intraperitoneal injection of 70mg/kg monocrotaline,pulmonary hypertension model was formed.The third and fourth groups were given traditional Chinese medicine prescription intragastric administration,5mL/day;the fifth group was given western medicine enalapril(0.05mg/day)intragastric administration;the first and second groups were given the same amount of normal saline intragastric administration.After 4 weeks of treatment,the levels of angiotensin-converting enzyme 2(ACE2),angiotensin 1-7(Ang1-7),phospholipase A2(PLA2)and serum amyloid protein A(SAA)in lung tissue were determined by enzyme-linked immunosorbent assay(ELISA).Results Compared with the model group,the levels of ACE2 and Ang1-7 in the high-dose,low-dose and enalapril groups were all decreased to varying degrees,with the most obvious reduction in the high-dose group and a slight decrease in the enalapril group.The results indicated that the combined treatment with traditional Chinese medicine could down-regulate the levels of ACE2 and Ang-1-7,dilate blood vessels and improve the pulmonary blood circulation.In addition,compared with the model group,the levels of inflammation markers PLA2 and SAA in the high-dose and low-dose groups were decreased to varying degrees,especially in the high-dose group.These results indicate that treatment can inhibit the release of inflammatory factors in vascular endothelial cells and play an anti-inflammatory role.Conclusion Shuangshen Gualou Decoction(双参瓜蒌汤)can down-regulate the levels of ACE2 and Ang1-7,play the role of vascular dilation,down-regulate the levels of inflammatory factors PLA2 and SAA,reduce the inflammatory apoptosis of vascular en
