摘  要:目的探究中药毛稔对伤口的愈合的治疗效果并探讨其潜在的作用机制。方法采取药物水煎法获取中药毛稔原液,按50倍、100倍、200倍、400倍稀释后作用于体外培养的人角质形成细胞HaCaT,研究分为对照(CON)组、实验(M50,M100,M200,M400)组,CCK8法检测细胞活力,划痕模型检验伤口愈合效果;液相色谱-质谱联用分析毛稔的代谢产物并构建药物-疾病-靶点相互作用网络预测中药毛稔干预伤口愈合的机制。结果与CON组相比,M50、M100、M200、M400组HaCaT细胞相对增殖率[(112.59±2.04)%、(119.04±2.15)%、(128.40±1.10)%、(133.72±2.83)%]增加(P<0.05)。M400组划痕愈合率在12 h和24 h[(86.93±4.90)%、(95.13±3.70)%]高于CON组[(36.29±2.97)%、(85.01±2.10)%],差异具有统计学意义(P<0.05)。非靶向代谢组学筛选中药毛稔有效活性成分539个,活性成分作用靶点与伤口愈合相交靶点592个。富集分析结果显示,交集靶点主要参与2655个生物过程,富集于92个细胞组分,主要涉及164个分子功能,并涉及180条信号通路。结论中药毛稔能够促进HaCaT细胞增殖和划痕损伤后愈合,可能通过亚麻酸甘油酯、白桦脂醇、L-色氨酸、桑黄素等活性成分作用于TNF、SRC、TP53等靶点达到治疗创伤的作用,其潜在机制可能与PI3K-Akt等信号通路有关,有望成为治疗创伤的有效药物。Objective To explore the therapeutic effect of Melastoma sanguineum Sims,a traditional Chinese medicine,on wound healing and explore its potential mechanism.Methods The crude liquid of Melastoma sanguineum Sims was obtained by drug decoction method,and it was diluted by 50 times,100 times,200 times and 400 times to act on human keratinocytes HaCaT cultured in vitro.The study included the control(CON)group and experimental(M50,M100,M200,M400)group.The cell viability was detected by CCK8 method,and the wound healing effect tested by scratch model.The metabolites of Melastoma sanguineum Sims were analyzed by liquid chromatography-mass spectrometry and a drug-disease-target interaction network was constructed to predict the mechanism of intervention of the drug in wound healing.Results As compared with the control group,the relative proliferation rates of HaCaT cells in M50,M100,M200 and M400 groups((112.59±2.04)%,(119.04±2.15)%,(128.40±1.10)%,(133.72±2.83)%)were significantly increased(P<0.05)in a concentration-dependent manner.The scratch healing rate of M400 group at 12 h and 24 h((86.93±4.90)%,(95.13±3.70)%)was higher than that of the control((36.29±2.97)%,(85.01±2.10)%),demonstrating statistically significant difference(P<0.05).A total of 539 effective active components were screened by non-targeted metabolomics and 592 targets of active components intersected with wound healing.The enrichment analysis showed that the intersection targets were mainly involved in 2655 biological processes,enriched in 92 cell components,mainly related to 164 molecular functions and 180 signaling pathways.Conclusion The traditional Chinese medicine,Melastoma sanguineum Sims,is expected to become an effective drug for the treatment of trauma because it can promote the proliferation of HaCaT cells and the healing after scratch injury.The drug might be used to treat trauma by acting on TNF,SRC,TP53 and other targets through active ingredients such as glyceryl linolenate,betulin,L-tryptophan,morin,etc.The potential mechanism of
