活性污泥中噬菌体的环境雌激素降解功能研究

摘　　要：环境雌激素(EEs)随污水排入自然水体,具有较大的生态环境风险和人体健康风险。采用宏基因组测序技术对普通活性污泥和双酚A(BPA)、邻苯二甲酸二丁酯(DBP)、壬基酚(NP)富集污泥中的噬菌体基因组进行分析发现,普通活性污泥和EEs富集污泥中可以鉴定的噬菌体均以管病毒科(Siphoviridae)、肌病毒科(Myoviridae)和足病毒科(Podoviridae)为主,但是不同污泥的噬菌体相对丰度组成存在显著差异。通过对噬菌体基因组进行功能基因注释发现,BPA富集污泥和NP富集污泥中管病毒科噬菌体分别有编码苯酚单加氧酶和脱氢酶的基因,有助于芳香族有机物矿化去除。噬菌体的细菌宿主分析发现,噬菌体广泛寄生于各类细菌,但主要是BPA富集污泥中有编码羟基化功能的苯酚单加氧酶基因的管病毒科噬菌体具有BPA降解功能,即具有重要的潜在辅助代谢EEs功能。As environmental estrogens(EEs)are discharged into natural water bodies along with wastewater,it will cause great risks to the ecological environment and human health.Using metagenomic sequencing technology to study ordinary activated sludge,as well as bisphenol A(BPA),dibutyl phthalate(DBP)and nonylphenol(NP)enriched sludges,it was found that the phages in ordinary activated sludge and EEs enriched sludges were mainly Siphoviridae,Myoviridae and Podoviridae detected.However,there were significant differences in the relative abundance composition of phages in different sludges.Through functional gene annotation of phage genomes,it was found that the phage genomes of Siphoviridae in BPA-and NP-enriched sludges encoded phenol monooxygenase and dehydrogenase,respectively.Monooxygenase and dehydrogenase would contribute to remove aromatic compounds by mineralization.By analyzing parasitic relationships between phages and bacteria,it was found that phages were widely parasitic in various bacteria.But it was Siphoviridae in BPA-enriched that had great potential auxiliary metabolic function for EEs,which encoded monooxygenase gene with BPA metabolic function.
