摘  要:‘冬枣’、枝条扭曲突变及其恢复突变是一套来源清晰的枣枝条扭曲变异材料。采用SSR标记鉴定,并通过形态与解剖结构观察揭示枝条扭曲变异的枝叶特征。37对SSR引物从‘冬枣’、枝条扭曲突变及其恢复直枝突变的基因组DNA扩增出52种带型,但未发现3种材料之间的差异。枝条扭曲突变及其恢复直枝突变的叶横径、叶周长、叶面积、长针刺、短针刺长度均显著小于‘冬枣’。恢复直枝突变的叶片厚度、上表皮厚度、栅栏组织厚度、栅栏组织厚度/海绵组织厚度、叶片组织结构紧实度均显著高于‘冬枣’和枝条扭曲突变。‘冬枣’叶片主叶脉厚度及枣吊导管面积、导管直径、导管壁厚度均显著大于枝条扭曲突变及其恢复直枝突变。‘冬枣’、枝条扭曲突变及其恢复直枝突变的遗传背景一致,枝叶性状差异突出,可用于枝条扭曲生长机制研究。‘Dongzao’(Ziziphus jujuba cv.‘Dongzao’),twisted branch mutant and its revertant are a set of mutation materials.The objective of this study was to identify these variants by SSR and explore the characteristics of leaf and branch of the twisted branch mutants based on morphological and anatomical observation.Fifty-two band types were amplified by 37 pairs of SSR primers from the genomic DNA of‘Dongzao’,twisted branch mutant and its revertant,but no differences among three materials were found.Leaf transverse diameter,leaf perimeter,leaf area,long thorn and short thorn length of twisted branch mutant and its revertant were significantly lower than those of‘Dongzao’.Leaf thickness,upper epidermis thickness,palisade tissue thickness,the ratio of palisade thickness/spongy tissue thickness and leaf tissue compactness of the revertant were significantly higher than those of‘Dongzao’and twisted branch mutant.Leaf main vein thickness,vessel area,vessel diameter and vessel wall thickness of‘Dongzao’were significantly larger than those of the twisted branch mutant and its revertant.The genetic background of‘Dongzao’,twisted branch mutant of‘Dongzao’and its revertant was the same,yet the branch and leaf traits distinctively varied in three materials.‘Dongzao’and both mutants could be used as materials for exploration of twisted branch growth mechanism.
