摘  要:鲢(Hypophthalmichthys molitrix)是中国特有主要淡水养殖鱼类之一,产量位居淡水鱼类第二位;但鲢喜跳跃,应激反应强烈,操作与运输较为不便。本研究的目的是确定苯唑卡因对鲢麻醉的合适浓度;并在合适浓度下探索鲢肝肾组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)酶活及部分免疫基因表达特征的影响。实验方法是将鲢放入不同浓度的苯唑卡因溶液中麻醉,确定合适的麻醉浓度,并测定此浓度下鲢肝脏和肾脏中总SOD和CAT活力的变化,以及IL-10、TNF-α、INF-γ、IgM等免疫基因表达的变化。结果表明:苯唑卡因麻醉鲢的合适浓度为28 mg/L。与对照组相比,鲢肝脏中的IL-10基因表达量在麻醉12 h和复苏6 h实验组中显著上升;TNF-α基因表达量在复苏1 h实验组中显著上升;INF-γ基因表达量分别在鲢麻醉6、9、24 h及复苏3 h和6 h实验组中显著变化;IgM基因表达量分别在鲢麻醉9、12、18、24 h及复苏6 h实验组中显著上升。鲢肾脏中的IL-10、TNF-α和INF-γ基因表达量在鲢麻醉组和复苏组中均显著下降;IgM基因表达量在鲢复苏3 h实验组中显著上升。鲢肝脏和肾脏中的T-SOD活力及CAT活力在麻醉组和复苏组中均有显著变化。在鲢肝脏和肾脏中,T-SOD活力呈先下降后上升趋势;在鲢肝脏中CAT活力呈先上升后下降趋势,在鲢肾脏中则呈现先下降后上升趋势,表明苯唑卡因对鲢不同组织酶活反应的特异性。本项研究为鲢麻醉运输、麻醉时鲢肝肾组织抗氧化酶酶活及部分免疫基因表达提供了理论依据。Hypophthalmichthys molitrix,a prominent freshwater aquaculture fish species native to China,holds the second position in terms of production among other freshwater fish.However,due to its pronounced response to stress conditions,such as jumping,the handling and transporting H.molitrix can be challenging and inconvenient.The fish were subjected to varying concentrations of benzocaine solution to ascertain the suitable concentration of benzocaine for inducing anesthesia in H.molitrix.Subsequently,alterations in the overall superoxide dismutase(SOD)and catalase(CAT)activity within the liver and kidney of H.molitrix were assessed,alongside variations in the expression of immunological genes,including IL-10,TNF-α,INF-γ,and IgM,to identify the optimal anesthetic concentration.The experimental findings revealed that a concentration of 28 mg/L of benzocaine was found to be suitable for inducing anesthesia in H.molitrix.Compared to the control group,the expression of IL-10 in the liver of H.molitrix exhibited a significant increase in the experimental groups subjected to 12 hours of anesthesia and 6 hours of resuscitation.Additionally,the expression of TNF-α showed a significant increase in the experimental group subjected to 1 hour of resuscitation.Furthermore,the expression of INF-γ demonstrated significant changes in the H.molitrix subjected to anesthesia for 6,9,and 24 hours and resuscitation for 3 and 6 hours,respectively.The expression of the IgM gene exhibited significant changes in the H.molitrix anesthesia groups at 6,9,and 24 hours,as well as in the resuscitation groups at 3 and 6 hours.Specifically,there was a significant increase in the expression of the IgM gene in the resuscitation groups at 9,12,18,24 hours,and 6 hours for H.molitrix anesthesia.Additionally,the expression of IL-10,TNF-α,and INF-γ in the kidney of H.molitrix showed a significant decrease in both the anesthesia and resuscitation groups.The expression of the IgM gene exhibited a significant increase in the experimental groups of H.molitr
