摘 要:【目的】本研究旨在通过建立团花绢野螟Diaphania glauculalis成虫触角转录组数据库,挖掘化学感受相关基因,为团花绢野螟成虫触角的化学感受机制及绿色防控技术提供理论基础。【方法】使用Illumina NovaSeq 6000平台对团花绢野螟成虫触角转录组进行测序,使用Trimmomatic和Trinity对测序所得的原始数据进行剪切、组装得到转录组数据;比对CDD,KOG,COG,NR,NT,PFAM和KEGG等数据库获得基因注释信息,筛选鉴定出化学感受相关基因;使用TPM(transcripts per million)评估化学感受相关基因的表达量;应用最大似然法构建团花绢野螟化学感受相关基因系统发育树。【结果】团花绢野螟雄成虫触角转录组测序获得52705124条unigenes,雌成虫触角转录组测序获得55391610条unigenes。通过与NR数据库比对注释,团花绢野螟成虫触角转录组注释unigenes共24192条,其中与亚洲玉米螟Ostrinia furnacalis的同源序列最多,有10679条。有14313条unigenes被注释到204289条GO功能条目,其中113387条unigenes注释到生物学进程,70115条unigenes注释到细胞组分,20787条unigenes注释到分子功能。有10721条unigenes注释到KOG数据库25个分类的11968功能条目。有12892条unigenes被注释到KEGG数据库5类代谢通路,归属于33个通路,其中注释到信号转导通路的unigenes最多,有1500条unigenes,进而筛选鉴定到136个化学感受相关基因,包括31个气味结合蛋白(odorant-binding protein,OBP)基因、24个化学感受蛋白(chemosensory protein,CSP)基因、50个气味受体(odorant receptor,OR)基因、20个离子型受体(ionotropic receptor,IR)基因、9个味觉受体(gustatory receptor,GR)基因和2个感觉神经元膜蛋白(sensory neuron membrane protein,SNMP)基因。【结论】本研究首次构建了团花绢野螟成虫触角转录组数据库,阐述了化学感受相关基因的种类和数量,为团花绢野螟化学感受基因功能分析及化学感受机制奠定分子基础,进�【Aim】This study aims to establish the adult antennal transcriptome database of Diaphania glauculalis and mine chemosensory-related genes,so as to provide a theoretical basis for the chemosensory mechanism of adult antenna of D.glauculalis,and green prevention and control technology.【Methods】The antennal transcriptome of D.glauculalis adults was sequenced using an Illumina NovaSeq 6000 platform.The raw data were edited using Trimmomatic and Trinity and assembled to obtain transcriptome data.The transcriptome data were compared to CDD,KOG,COG,NR,NT,PFAM and KEGG databases to obtain gene annotation information and identify the chemosensory-related genes.The expression levels of the chemosensory-related genes were assessed using TPM(transcripts per million).The phylogenetic tree of the chemosensory-related genes of D.glauculalis was constructed using the maximum likelihood method.【Results】A total of 52705124 unigenes were obtained from the male adult antennal transcriptome of D.glauculalis and 55391610 unigenes from the female adult antennal transcriptome.By comparing the annotation with NR database,24192 unigenes were annotated,among them,the most(10679)homologous sequences were from Ostrinia furnacalis.There were 14313 unigenes annotated to 204289 GO functional entries,including 113387 unigenes annotated to biological processes,70115 unigenes annotated to cellular components,and 20787 unigenes annotated to molecular functions.There were 10721 unigenes annotated to 11968 functional entries in 25 classifications of the KOG database.There were 12892 unigenes annotated to five categories of metabolic pathways in the KEGG database,attributed to 33 pathways,with the largest number of unigenes(1500)annotated to signal transduction pathways.A total of 136 chemosensory-related genes were identified,including 31 odorant-binding protein(OBP)genes,24 chemosensory protein(CSP)genes,50 odor receptor(OR)genes,20 ionotropic receptor(IR)genes,nine gustatory receptor(GR)genes and two sensory neuron membrane protein(SNMP)
