摘 要:CRISPR关联基因12f(Cas12f)由于蛋白序列短,便于基因递送及后续的基因治疗,受到广泛的关注及越来越多的研究报道。本文系统性地回顾和总结了基因编辑系统CRISPR(成簇的规则间隔短回文重复序列)/Cas12f及其衍生的相关基因编辑工具的研究进展,深入探讨了Cas12f的结构特性、间隔子相邻基序(PAM)识别与切割机制,以及基于CRISPR/Cas12f的碱基编辑和转录调控,以期为进一步开发基于Cas12f的基因编辑及治疗工具提供参考。CRISPR-associated 12f(Cas12f),known for its short protein sequence which facilitates gene delivery and subsequent gene therapy,has attracted widespread attention and an increasing number of research reports.This article systematically reviews the research progress of the gene editing system CRISPR(clustered regularly interspaced shortpalindromic repeats)/Cas12f and its derived gene editing tools including the structural features,protospacer adjacent motif(PAM)recognition and cutting mechanisms of Cas12f,as well as base editing and transcriptional regulation based on CRISPR/Cas12f,with the aim of providing references for further development of gene editing and therapeutic tools based on Cas12f.
