摘  要:基于AgilentPoroshellEC-C18色谱柱(4.6 mm×100 mm,2.7μm),作者研究了提高盐酸莫西沙星及其对映异构体分离度的方法。结果表明,当流动相A为含有醋酸铵、硫酸铜和L-异亮氨酸的水溶液,且醋酸铵浓度为50 mmol·L^(-1),硫酸铜的浓度为5 mmol·L^(-1),L-异亮氨酸的浓度为10 mmol·L^(-1),流动相B为甲醇,流动相A与流动相B体积之比为70∶30,流速为1.0 m L·min^(-1),进样量为10μL,柱温为40℃,检测波长为293 nm时,盐酸莫西沙星对映异构体分离度达到6.4。该方法不使用手性色谱柱,相比现有技术,可以更好地分离盐酸莫西沙星及其对映异构体。Based on the Agilent Poroshell EC-C18 chromatography column(4.6 mm×100 mm,2.7μm),the authors studied a method to improve the separation of moxifloxacin hydrochloride and its enantiomers.The results showed that when mobile phase A was an aqueous solution containing ammonium acetate,copper sulfate,and L-isoleucine,with ammonium acetate concentration of 50 mmol·L^(-1),copper sulfate concentration of 5 mmol·L^(-1),L-isoleucine concentration of 10 mmol·L^(-1),mobile phase B was methanol,the volume ratio of mobile phase A to mobile phase B was 70∶30,the flow rate was 1.0 mL·min^(-1),the injection amount was 10μL,the column temperature was 40℃,and the detection wavelength was 293 nm,the resolution of moxifloxacin hydrochloride isomers reached 6.4.This method did not use chiral chromatography columns,and had a better separation moxifloxacin hydrochloride and its isomers compared to existing technologies.
