摘  要:[目的]建立1种牛病毒性腹泻病毒(BVDV)与冠状病毒(BCoV)混合感染的PCR诊断方法。[方法]选择山东省某奶牛场内发生BVDV和BCoV混合感染的98头奶牛作为研究对象,采集其粪便和组织脏器,提取病毒基因组RNA,经反转录后,设计引物,进行PCR检测,分析检测情况及特异性、重复性。[结果]BVDV和BCoV混合感染病例40份(40.82%),其中粪便样本33份(40.27%),组织脏器样本7份(43.75%)。BVDV单一感染病例27份(27.55%),其中粪便样本21份(25.61%),组织脏器样本6份(37.50%)。BCoV单一感染病例32份(32.65%),其中粪便样本28份(34.15%),组织脏器样本4份(25.00%)。[结论]本研究中BVDV和BCoV混合感染的PCR诊断方法准确,具有良好的特异性和重复性,为今后临床正确诊断这两种病毒提供参考,也为后续精准科学防控奠定基础。[Objective]To establish a PCR diagnostic method for mixed infection of Bovine Viral Diarrhea Virus(BVDV) and Bovine Coronavirus(BCoV).[Method]98 cows infected with BVDV and BCoV in a dairy farm in Shandong were selected as the research subjects.Their feces and tissues were collected and virus genomic RNA was extracted.After reverse transcription,primers were designed for PCR detection,and the detection status,specificity and repeatability were analyzed.[Result] 40 cases were infected with BVDV and BCoV(40.82%),including 33 fecal samples(40.27%) and 7 tissue samples accounted for 7 cases(43.75%).There were 27 cases(27.55%) of BVDV single infection,including 21 fecal samples(25.61%) and 6 tissue samples(37.50%).There were 32 cases(32.65%) of BCoV single infection,including 28 fecal samples(34.15%)and 4 tissue samples(25.00%).[Conclusion]In this study,the PCR diagnostic method for the mixed infection of BVDV and BCoV was accurate,with good specificity and repeatability,which provided a reference for the correct diagnosis of these two viruses in clinical practice and laid a foundation for precise and scientific prevention and control in the future.
