摘 要:目的 对乌鲁木齐市结核分枝杆菌进行群体遗传学研究,揭示其遗传变异情况、单倍型分布和群体历史动态。方法 使用全基因组测序技术对乌鲁木齐市七区一县的结核分枝杆菌群体进行测序,通过单核苷酸多态性进行基因分型,开展群体遗传多样性、遗传结构、基因流动和历史动态分析。结果 结核分枝杆菌群体样本共457个,其中检测出22个单倍型,单倍型多样性(H_(d))为0.890 49,核苷酸多态性(π)为0.496 83,群体遗传多样性较高。AMOVA分析显示,结核分枝杆菌群体遗传变异主要发生在群体内,群体间遗传分化系数(F_(st))为0.068 9,基因流(N_(m))为3.42,结核分枝杆菌群体间有较高的基因流动。中性检验和错配分析表明乌鲁木齐市结核分枝杆菌群体可能没有经历明显的快速扩张事件。结论 乌鲁木齐市结核分枝杆菌群体遗传性较高,主要发生群体内遗传变异,具有明显基因流动。Objective Population genetic study of mycobacterium tuberculosis in Urumqi to reveal genetic variation,haplotype distribution and population history dynamics.Methods Whole genome sequencing technology was used to sequence mycobacterium tuberculosis populations from seven 7 districts and 1 one county in Urumqi,genotyping by single nucleotide polymorphisms,and carrying out analyses of population genetic diversity,genetic structure,gene flow,and historical dynamics.Results A total of 457 mycobacterium tuberculosis population samples were collected,of which 22 haplotypes were detected,with a haplotype diversity(H_d) of 0.890 49 and nucleotide polymorphism(π) of 0.496 83,resulting in a high genetic diversity of the population.AMOVA analysis showed that genetic variation in the mycobacterium tuberculosis population occurred mainly within the population,with a coefficient of genetic differentiation between populations(F_(st)) of 0.068 9 and a gene flow(N_m) of 3.42,and that there was a high level of gene flow between mycobacterium tuberculosis populations.Neutrality tests and mismatch analyses is suggested that the mycobacterium tuberculosis population in Urumqi may not have experienced a significant rapid expansion event.Conclusion Mycobacterium tuberculosis populations in Urumqi have high heritability and mainly undergo intra-population genetic variation with significant gene flow.
