摘  要:目的:通过社交隔离法结合行为学、宏观表征、客观病理指标以及方剂反证法建立并评价小鼠阴虚火旺证抑郁症模型。方法:采用5周的社交隔离法建立小鼠抑郁症模型,通过宏观表征、舌爪红色度r值、颈动脉血管及血清甲状腺素含量进行证候判别,同时予以滋水清肝饮反证病证结合模型的可靠性。结果:与空白组比较,造模后模型小鼠出现精神倦怠、反应迟缓、有攻击行为、形体消瘦、毛发散乱等表现,模型组小鼠蔗糖偏好率、进入旷场中心次数及时间均显著降低(P<0.05),悬尾不动时间增加(P<0.01),抑郁症模型构建成功;同时,模型组小鼠进食量及饮水量均显著增加(P<0.01,P<0.05);舌r值、前后爪r值显著升高(P<0.05,P<0.01),颈动脉舒张期和收缩期血管径均显著降低(P<0.01,P<0.05),心率显著升高(P<0.05),且伴有血中甲状腺素含量增加(P<0.01)。而滋水清肝饮较模型组有显著的治疗调节作用(P<0.01,P<0.05)。结论:持续5周的社交隔离法造模可成功制备阴虚火旺证抑郁症小鼠模型。Objective:The depression model of yin deficiency and fire effulgence syndrome was established by social isolation combined with behavioral,macroscopic and objective pathological indicators and prescription counterevidence mothod.Methods:A mouse depression model was established by social isolation for 5 weeks,and the syndrome was identified by macroscopic characteristics,redness of tongue and paw,carotid artery blood vessels and serum thyroxine content.Then Zishui Qinggan Yin was used to counter the reliability of the desease-syndrome combination model.Results:Compared with the control group,the model mice showed mental burnout,slow reaction,aggressive behavior,emaciation,and loose hair after modeling,the sucrose preference rate,the number and time of entering the open field center in the model group were significantly decreased(P<0.05),and the tail suspension immobility time was increased(P<0.01),indicating that the depression model was successfully established.In addition,the consumption of food and water in the model group were significantly increased(P<0.01,P<0.05).The r values of tongue and paw color were significantly increased(P<0.05,P<0.01),the diastolic and systolic diameters of carotid artery were significantly decreased(P<0.01,P<0.05),the heart rate was significantly increased(P<0.05),and the content of thyroxine in blood was increased(P<0.01).After the administration of Zishui Qinggan Yin,the above indicators were all reduced to varying degrees than the model group(P<0.01,P<0.05).Conclusion:The depression mouse model of yin deficiency and fire effulgence can be successfully established by social isolationfor5weeks.
