摘 要:目的:探讨淫羊藿和仙茅药对(EBCOs)在长链非编码RNA(lncRNA)水平上对高浓度葡萄糖诱导的成骨细胞损伤的保护机制。方法:淫羊藿干叶和仙茅干根茎热水提取制备EBCOs;成骨细胞分为对照组(1.65 mmol/L葡萄糖处理24 h)、HG模型组(33.1 mmol/L葡萄糖处理24 h)和EBCO组(33.1 mmol/L葡萄糖处理24 h并用10μg/mL含EBCOs血清干预);lncRNA测序检测lncRNA表达水平;细胞转染沉默lncRNA Gabpb1-AS1;qRT-PCR检测lncRNA Gabpb1-AS1和Nrf2 mRNA表达量;Western Blot检测Keap1、Nrf2和BMP2蛋白表达量。结果:HG组较control组lncRNA Gabpb1-AS1表达显著增加(P<0.01),EBCOs干预后可显著降低Gabpb1-AS1表达(P<0.01)。转染沉默lncRNA Gabpb1-AS1后,lncRNA Gabpb1-AS1显著降低(P<0.01),Nrf2和BMP2蛋白表达显著增加(P<0.01),Keap1蛋白表达显著降低(P<0.01)。结论:EBCOs可通过下调lncRNA Gabpb1-AS1来减轻高糖诱导的成骨细胞损伤。Objective:To investigate the protective mechanism of Epimedi Folium and Curculiginis Rhizoma(EBCOs)against osteoblast injury induced by high glucose concentration at the IncRNA level.Methods:EBCOs was extracted from dried leaves of Epimedi Folium and dried rhizomes of Curculiginis Rhizoma.Osteoblasts were divided into three groups:control group(1.65 mmol/L glucose treatment for 24 h),HG model group(33.1 mmol/L glucose treatment for 24 h)and EBCO group(33.1 mmol/L glucose treatment for 24 h and 10μg/mL EBCOs-containing serum intervention).The IncRNA expression level was detected by IncRNA sequencing.Expressions of IncRNA Gabpb1-AS1 and Nrf2 mRNA were detected by qRT-PCR.Cell transfection silenced IncRNA Gabpb1-AS1.The protein expression of Keapl,Nrf2 and BMP2 were detected by Western Blot.Results:The IncRNA Gabpb1-AS1 in HG group was significantly increased compared with control group(P<0.01),and decreased after EBCOs intervention(P<0.01).After silencing the IncRNA Gabpb1-AS1 by transfection,the IncRNA Gabpb1-AS1 was significantly decreased(P<0.01),the protein expressions of Nrf2 and BMP2 were significantly increased while the protein expressions of Keapl were significantly decreased(P<0.01).Conclusion:EBCOs may relief high glucose-induced damage in osteoblastsbydown-regulating IncRNAGabpb1-AS1.
