摘  要:目的:采用网络药理学和体内试验研究守正代茶饮对正常小鼠免疫增强的潜在作用机制。方法:通过TCMSP、Swiss Target Prediction数据库获取守正代茶饮的活性成分及对应靶标;GeneCards数据库得到免疫相关靶点并与药物靶点取交集;通过STRING和DAVID数据库,对交集靶点进行PPI网络分析和GO、KEGG富集分析,于Cytoscape 3.9.1构建“守正代茶饮-潜在活性化合物-靶点”和“成分-靶点-通路”网络,AutoDock Tools进行分子对接验证。小鼠碳廓清试验、迟发型变态反应试验、血清溶血素试验及Western Blot对部分靶点进行验证。结果:守正代茶饮的活性成分108种,增强免疫力交集靶点114个,核心成分为槲皮素、β-谷甾醇、山柰酚、豆甾醇、木犀草素等;关键靶点为PTGS2、HSP90AB1、PRKACA、RELA、Akt1等;主要参与药物的应答、基因表达的正调节、凋亡过程的负调控、TNF、PI3K-Akt、IL-17等生物过程和信号通路;关键靶点和核心成分亲和力较强。动物试验结果显示,与空白组比较,守正代茶饮低、中、高剂量组增强小鼠迟发型变态反应水平;中、高剂量组改善巨噬细胞吞噬活性;中、高剂量组p-PI3K、p-Akt、p-P65蛋白表达均上调(P<0.01)。结论:守正代茶饮具有增强免疫力的功能,可能通过PI3K-Akt/NF-κB信号通路促进免疫活性物质的释放,发挥免疫调节作用。Objective:Network pharmacology and in vivo experiments were used to investigate the potential mechanism of Shouzheng drink as tea to immune enhancement in normal mice.Methods:By obtaining the active ingredients and corresponding target of TCMSP,Swiss Target Prediction databases,established the data sets;GeneCards database obtains immune-related targets and intersects with drug targets;PPI network analysis and GO and KEGG enrichment analysis of the intersection targets by STRING and DAVID databases,The network map of"Shouzheng drink as tea-potential active compound-target"and"component-target-pathway"network map were constructed by Cytoscape 3.9.1,molecular docking validation using AutoDock Tools.Mice carbon clearing experiment,delayed type hypersensitivity experiment and serum hemolysin experiment and Western Blot to verify some targets.Results:A total of 108 active ingredients of Shouzheng drink as tea were obtained,and 114 intersection targets of the immunity enhancement,The core ingredients of Shouzheng drink as tea in the enhancement of immunity included quercetin,beta-sitosterol,kaempferol,stigmasterol,luteolin;the core targets included PTGS2,HSP90AB1,PRKACA,RELA,Akt1,it is mainly involved in the response to drugs,positive regulation of gene expression,and negative regulation of the apoptosis process,TNF,PI3K-Akt and IL-17;key targets and core components have a strong affinity.Results of the animal experiments are shown,compared with the blank group,the low,medium and high doses groups of Shouzheng drink as tea enhanced the level of delayed allergic reaction in mice,and the medium and high doses groups could improve the phagocytic index of macrophage,the protein expression of p-PI3K,p-Akt and p-P65 were up regulated in the medium and high-dose Shouzheng drink as tea groups(P<0.01).Conclusion:Shouzheng drink as tea has the function of enhancing immunity,perhaps through PI3K-Akt/NF-κB signaling pathway promotes the release of immune active substances and exerts immune regulatory effects.
