祛瘀解毒方对子宫内膜异位症大鼠在位子宫内膜铁自噬的影响

摘　　要：目的观察祛瘀解毒方对子宫内膜异位症(EMS)模型大鼠在位子宫内膜组织铁自噬的影响,探讨其治疗EMS的作用机制。方法50只雌性SD大鼠随机分为空白组、模型组、西药组和祛瘀解毒方高、低剂量组,每组10只。除空白组外,其余组均采用自体移植法造模,成模后分别予相应溶液灌胃4周。HE染色观察在位子宫内膜组织形态,试剂盒检测在位子宫内膜组织亚铁离子(Fe2+)含量,Westernblot检测在位子宫内膜组织核受体共激活因子4(NCOA4)、铁蛋白重链1(FTH1)、LC3BⅡ/Ⅰ、p62蛋白表达,qPCR检测在位子宫内膜组织NCOA4、FTH1、LC3B、p62 mRNA表达,免疫荧光共定位检测在位子宫内膜组织NCOA4和LC3B共定位情况。结果与空白组比较,模型组大鼠在位子宫内膜组织Fe2+含量降低(P<0.01),NCOA4蛋白表达及LC3BⅡ/Ⅰ比值降低(P<0.01),FTH1、p62蛋白表达升高(P<0.05,P<0.01),NCOA4 mRNA表达降低(P<0.01),p62mRNA表达升高(P<0.01),NCOA4-LC3B共定位减少(P<0.01);与模型组比较,各给药组在位子宫内膜组织Fe2+含量升高(P<0.05,P<0.01),西药组FTH1、p62蛋白表达降低(P<0.05,P<0.01),祛瘀解毒方高剂量组LC3BⅡ/Ⅰ比值升高(P<0.01),FTH1、p62蛋白表达降低(P<0.05,P<0.01),祛瘀解毒方低剂量组NCOA4蛋白表达及LC3BⅡ/Ⅰ比值升高(P<0.05),p62蛋白表达降低(P<0.05),各给药组NCOA4mRNA表达升高(P<0.01),NCOA4-LC3B共定位增加(P<0.05,P<0.01)。各组大鼠在位子宫内膜组织形态未见明显异常。结论祛瘀解毒方可能通过激活NCOA4介导的铁自噬通路恢复EMS大鼠在位内膜铁蛋白活性,上调铁离子含量,纠正铁稳态失衡,从而抑制EMS发展。Objective To investigate the effects of Quyu Jiedu Prescription on iron autophagy in eutopic endometrial tissue of rats with endometriosis(EMS);To explore its mechanism in treating EMS.Methods A total of 50 female SD rats were randomly divided into blank group,model group,Western medicine group,Quyu Jiedu Prescription high-and low-dosage groups,with 10 rats in each group.Except for the blank group,all other groups were modeled using autologous transplantation method,and corresponding solutions were given by gavage for 4 weeks.HE staining was used to observe the morphology of eutopic endometrial tissue,the reagent kit was used to detect the content of ferrous ions(Fe2+)in eutopic endometrial tissue,Western blot was used to detect the protein expressions of nuclear receptor coactivator factor 4(NCOA4),ferritin heavy chain 1(FTH1),LC3BⅡ/Ⅰand p62 in eutopic endometrial tissue,the mRNA expressions of NCOA 4,FTH1,LC3B and p62 were detected by qPCR,immunofluorescence colocalization was used to detect the co-localization of NCOA4 and LC3B in eutopic endometrial tissue.Results Compared with the blank group,the content of Fe2+in eutopic endometrial tissue of the model group decreased(P<0.01),the expression of NCOA4 protein and LC3BⅡ/Ⅰratio decreased(P<0.01),the expressions of FTH1 and p62 protein increased(P<0.05,P<0.01),the expression of NCOA4 mRNA decreased(P<0.01),the expression of p62 mRNA increased(P<0.01),and the co-localization of NCOA4-LC3B decreased(P<0.01).Compared with the model group,the content of Fe2+in eutopic endometrial tissue of each administration group increased(P<0.05,P<0.01),the expressions of FTH1 and p62 protein in Western medicine group decreased(P<0.05,P<0.01),the ratio of LC3BⅡ/Ⅰin Quyu Jiedu Prescription high-dosage group increased(P<0.01),the expression of FTH1 and p62 protein decreased(P<0.05,P<0.01),the expression of NCOA4 protein and LC3BⅡ/Ⅰratio increased in Quyu Jiedu Prescription low-dosage group(P<0.05),while the expression of p62 protein decreased(P<0.05),the expression
