摘  要:虎红是一种阴离子型生物染色素,在酸性条件下游离的羧基可以和药物分子中的羟基通过氢键相互作用形成超分子体系,用于共振光散射光谱法的定量分析。本实验以氯霉素为应用对象,详细考察了实验条件对虎红-氯霉素超分子体系共振光散射强度的影响,获得最佳测试条件:虎红溶液浓度为4.9×10^(-4)mol·L^(-1),体积1.5 mL,盐酸体积1.0 mL,氯霉素浓度和体积分别为100μg·mL^(-1)和1.0 mL。方法学考察显示,氯霉素浓度在0-10μg·mL^(-1)范围内线性良好(y=3343.5x+21.857,R^(2)=0.9958),检出限为0.0015μg·mL^(-1),氯霉素加标样品2.0、2.5、3.0μg·mL^(-1)准确度考察,平均回收率分别为100.4%、100.5%和100.3%,相对标准偏差分别为1.4%、1.1%和1.9%(n=5)。该方法用于氯霉素滴眼液实际样品含量测定,获得标示量百分含量平均值为100.0%,RSD%为1.9%(n=6)。反应机理研究表明氯霉素和虎红的最佳结合摩尔比为1:2(mol/mol)。上述结果表明本实验开发的基于虎红超分子体系的共振光散射光谱法为氯霉素定量分析提供一种含量测定方法。Rose bengal is an anionic biochromatin.Under acidic conditions,the free carboxyl group of rose bengal can interact with the hydroxyl group of drug to form a supermolecule system through hydrogen bonding,which is used for quantitative analysis by reso-nance light scattering spectrometry.In this experiment,chloramphenicol was used as the application object,and the influence of experimental conditions on the resonance light scattering intensity of rose bengal-chloramphenicol supermolecule system was investiga-ted in detail.The best detection conditions were obtained:rose bengal solution concentration was 4.9x10^(-4) mol ^(-1),volume was 1.5 mL,hydrochloric acid volume was 1.0 mL,chloramphenicol concentration and volume were 100μg:mL^(-1)and 1.0 mL.Meth-odological investigation showed that good linear correlation was obtained at chloramphenicol concentration of O-10μg·mL^(-1)(y=3343.5x+21.857,R^(2)=0.9958),with a detection limit of 0.0015μg·mL^(-1).The accuracy was evaluated at the chloramphenicol spiked sample of 2.0,2.5,3.0μg mL',with average recoveries of 100.4%,100.5%,and 100.3%,and relative standard devia tions of 1.4%,1.1%,and 1.9%(n=5),respectively.This method was applied to the determination of the actual sample content of Chloramphenicol eye drops,and the average content of the labeled percentage was 100.0%,RSD%was 1.9%(n=6).The study of reaction mechanism showed that the optimal molar ratio of chloramphenicol to rose bengal was 1:2(mol/mol).The above results indicate that the resonance light scattering spectrometry based on rose bengal supermolecule system developed in this experiment provides a new method for quantitative analysis of chloramphenicol.
