怡肾丸对单侧输尿管结扎模型大鼠LC3及Beclin-1表达的影响

摘  要:目的:探讨怡肾丸对单侧输尿管结扎模型大鼠肾脏LC3及Beclin-1表达的影响。方法:将64只雄性SD大鼠随机分为假手术组(16只)、UUO模型组(16只)、UUO模型+怡肾丸组(16只)、UUO模型+依那普利组(16只)。手术组大鼠游离并结扎左侧输尿管制备肾纤维化模型,假手术组只游离不结扎左侧输尿管。各给药组于造模当日开始给予相应药物,假手术组和模型组灌胃等体积蒸馏水,连续灌胃3周。于术后第3、7、21天分批处死各组大鼠,取血清及左肾标本,检测血清肌酐、尿素氮,HE染色、Masson染色观察其病理形态学改变,免疫组织化学法及qRT-PCR法分别检测肾组织LC3及Beclin-1的蛋白及mRNA表达水平。结果:与假手术组相比,模型组血肌酐、尿素水平均升高(P<0.01);与模型组相比,治疗组肾功能上升速度缓慢(P<0.05);假手术组大鼠肾脏未见明显病理改变。模型组大鼠肾小球硬化,肾小管高度扩张萎缩,小管基底膜变薄,间质区细胞大量增生及纤维化,并随梗阻时间延长而加重,治疗组病理损伤明显减轻,胶原纤维沉积减少(P<0.05);与假手术组相比,模型组的LC3、Beclin1水平升高(P<0.05);与模型组相比,治疗组的LC3、Beclin1表达水平显著上升(P<0.05)。结论:UUO大鼠肾损害过程中自噬被激活,而怡肾丸增强了LC3、Beclin-1自噬相关蛋白的表达水平,并减轻肾纤维化以保护肾脏。Objective:To explore the effect of Yishen Pill on the expression of LC3 and Beclin-1 in the kidney of unilateral ureteral ligation model rats.Methods:64 male SD rats were randomly divided into sham-operated group(16 rats),UUO model group(16 rats),UUO model with Yishen pill group(16 rats),and UUO model with Enalapril group(16 rats).The renal fibrosis model was prepared by freeing and ligating the left ureter in the surgical group,and only the left ureter was free and not ligated in the sham-operated group.Each drug administration group was given the corresponding drug on the day of modeling,and the sham-operated group and the model group were gavaged with an equal volume of distilled water for 3 weeks.The serum and left kidney specimens were collected on postoperative days 3,7 and 21,and serum creatinine and urea nitrogen were measured,and the pathomorphological changes were observed by HE staining and Masson staining.Results:Compared with the sham-operated group,the blood creatinine and urea levels were increased in the model group(P<0.01);compared with the model group,the kidney function in the treated group increased slowly(P<0.05);no significant pathological changes were seen in the kidneys of the rats in the sham-operated group.In the model group,the glomeruli were sclerotic,the tubules were highly dilated and atrophied,the tubular basement membrane was thinned,and the cells in the interstitial area were heavily proliferated and fibrotic,which worsened with the prolongation of the obstruction time;in the treatment group,the pathological damage was significantly reduced and collagen fiber deposition was decreased(P<0.05);compared with the sham-operated group,the levels of LC3 and Beclin1 in the model group were increased(P<0.05);compared with the model group,the levels of The expression levels of LC3 and Beclin1 were significantly upregulated in the treatment group compared with the model group(P<0.05).Conclusion:Autophagy was activated during renal damage in UUO rats,while Yishen pill enhanced the expression
