摘  要:目的:优选骨痹合剂的纯化工艺。方法:以纯化后出膏率以及有效成分青藤碱、芍药苷、β-蜕皮甾酮的转移率为指标,分别考察碱沉淀法、平板膜超滤法和醇沉法对骨痹合剂原复方的纯化效果,以筛选最优的纯化方法。结果:在3种纯化工艺中,醇沉法纯化效果最好,当提取液浓缩至含生药量为1.9 g/mL,含醇量为80%,加醇体积为浓缩液体积4倍量时,出膏率可减少至10.28%,青藤碱、芍药苷、β-蜕皮甾酮3种有效成分的转移率分别为94.21%、92.86%、90.80%。结论:采用醇沉法对骨痹合剂纯化效果最好,该法操作简单、工艺稳定,可为制剂进一步开发提供参考。Objective:To optimize the purification technology of Gubi Mixture.Methods:Taking the paste yield and the transfer rate of the active ingredients sinomenine,paeoniflorin and β-ecdysterone as indexes,the purification effect of alkali precipitation method,plate membrane ultrafiltration method and alcohol precipitation method on Gubi mixture original compound were investigated respectively in order to screen the best purification method.Results:Alcohol precipitation method has the best purification effect among the three purification processes,When the decoction is concentrated to 1.9 μg/mL of raw herbs content,80% of alcohol content,and the volume of alcohol added is four times the volume of concentrated liquid,the paste yield was 10.28%,And the transfer rates of sinomenine,paeoniflorin and β-ecdysterone were 94.21%、92.86% and 90.80%.Conclusion:Gubi mixture has the best purification effect by alcohol sedimentation method,This method is simple and stable,which can provide a reference for the next development of drug preparation.
