摘 要:目的:建立测定蓝芩杂多糖含量的苯酚硫酸法,并研究其抗氧化能力以及细胞增殖活力,为后期开发利用提供理论支持。方法:用苯酚硫酸法测定蓝芩杂多糖含量,对最大吸收波长、苯酚质量分数等因素进行优化,并进行方法学考察,随后利用不同体积分数乙醇沉淀得到4个级分LD20、LD40、LD60和LD80。通过DPPH自由基清除率和CCK-8法来对抗氧化活性、免疫活性,并进行评价。结果:检测方法的最佳条件:6%苯酚溶液0.8 mL,硫酸4.5 mL,40℃水浴20 min,测得蓝芩总多糖含量为72.04%。LD20、LD40、LD60、LD80均表现出明显的抗氧化活性,除LD80外都能不同程度促进巨噬细胞增殖。结论:蓝芩多糖有较强抗氧化能力和细胞增殖活力,研究结果为进一步深入开发新型产品提供了数据支撑。Objective:A phenol-sulfuric acid method was established to determine the content of polysaccharide in Lanqin,and the antioxidant capacity and cell proliferation activity were studied to provide solid theoretical support for later development and utilization.Methods:The phenolic acid method was used to determine the content of polysaccharide in Lanqin,and the maximum absorption wavelength,phenol mass fraction and other factors were optimized,and the methodology was investigated,and then four fractions LD20,LD40,LD60 and LD80 were obtained by precipitation with different volume fractions of ethanol.Antioxidative activity and immune activity were evaluated by DPPH free radical scavenging rate and CCK-8 method.Results:The optimal conditions of the detection method:0.8mL of 6% phenol solution,4.5 mL of sulfuric acid,40℃ water bath for 20 min,and the total polysaccharide content of Lanqin was measured to be 72.04%.LD20,LD40,LD60,and LD80 all showed obvious antioxidant activities,and all of them except LD80 could promote the proliferation of macrophages to varying degrees.Conclusion:Lanqin polysaccharide has strong antioxidant capacity and cell proliferation activity,which lays the foundation for its further in-depth research and development into new products.
