摘 要:目的:探索白果外种皮中银杏酚酸的中试生产调试及其稳定性试验研究。方法:在确定白果外种皮中银杏酚酸的最佳提取参数、有机溶剂萃取纯化参数以及大孔树脂柱层析纯化参数后完成中试调试,并进行银杏酚酸的稳定性研究。结果:在确定的白果外种皮中银杏酚酸提取纯化工艺下,平均每批烘干白果外种皮投料量为20 kg,可以获得银杏酚酸2.43 kg,产率为11.98%,银杏酚酸含量为68.38%,回收率为75.76%。在参照2020版《中国药典》对提纯产品进行了银杏酚酸的稳定性试验,确定银杏酚酸的包装方式为密封避光保存,首先采用圆底封盖塑料管密封包装,再用自封袋进行二次包装,最后在最外层采用塑封袋进行包装。由于长期试验时限不足,目前初步得出该包装下银杏酚酸在6个月内含量稳定。结论:该研究将白果外种皮中银杏酚酸的回收利用生产工艺进行中试调试并对银杏酚酸的稳定性进行研究,为银杏产业中银杏酚酸的产业化实施提供参考数据。Objective:Exploring the pilot production debugging and stability test of ginkgo phenolic acid in Ginkgo extra seed bark.Methods:After determining the best extraction parameters of ginkgo phenolic acid,organic solvent extraction,and purification parameters of large porous resin column chromatography,the pilot test was completed,and the stability of ginkgo phenolic acid was studied.Results:Under the purification process of phenolic acid of Ginkgo biloba,the average feeding amount of each batch of dried Ginkgo external seed skin is 20 kg,and 2.43kg of ginkgo phenolic acid can be obtained,the yield is 11.98%,the content of ginkgo phenolic acid is 68.38%,and the recovery rate is 75.76%.According to the 2020 edition of Chinese Pharmacopoeia,the stability test of the phenolic acid of Ginkgo is conducted.It is determined that the packaging method of ginkgo phenolic acid is to save from light.First,the plastic pipe with round bottom cover is used,and then the second packaging with self-sealing bag,and finally,the plastic sealing bag is used in the outermost layer.Due to the insufficient long-term test time limit,it is preliminarily concluded that the content of ginkgo phenolic acid under this package is stable within 6 months.Conclusion:In this study,the production process of ginkgo phenolic acid in the seed skin of Ginkgo was tested and the stability of ginkgo phenolic acid was studied,which provided reference data for the industrialization implementation of ginkgo phenolic acid in Ginkgo industry.
